The Rhubarb Zhechong Pill S <sub> 180 </ Sub> Tumor-bearing Mouse Tumor Cell Apoptosis,

Purpose:To Explores the mechanism of antitumor effect of Rhubarb zhi bug pill,and then provide the theoretical basis as its clinical application and production of new medicine ,by Choosing a tumor burdened mice as the research object,observing the role of Rhubarb zhi bug pill to inhibit tumor and induce apoptosis.Material and method:Establishing the animal model of crowe's (S180)sarcoma as the mice model,dividing tumor-burdened mice into Rhubarb zhi bug pill high dose group, medium dose group and low dose group and giving them with Rhubarb zhi bug pill of its 82g/kg, 41g/kg, 20g/kg doses continually for 10 days, meanwhile setting (models) negative control group, the normal group and western medicine (cyclophosphamide) control group, observating the rate of tumor suppressor ; Using flow cytometric to analysis the apoptosis rate of tumor cells;Using electron microscope to observe the ultrastructure of tumor cells;Using situ hybridization method to detect the expression of Survivin .Results:(1) The inhibition rate to the tumor-burdened tumors in the mice of Rhubarb zhi bug pill high dose group, medium dose group and low dose group to 50.00%, 41.95% and 31.03% respectively ,there is significant difference compared with model group (P<0.05, P<0.01), especially with Rhubarb zhi bug pill high-dosage group obviously;(2) Under a optical microscope, the tumor areas and the tumor cell nests are observed to be decreased in the mice of Rhubarb zhi bug pill group and the numbers of tumor cell are reduced obviously;Under a transmission electron microscope tumor cells volumes in the mice of Rhubarb zhi bug pill group are decreased significantly, the microvillis of cell surface are reduced even absently, and can be seen apoptotic bodies;(3) It is proved that the apoptotic cells all can be seen in the tumor tissue of Rhubarb zhi bug pill treatment groups using flow cytometry;(4) Each of Rhubarb zhi bug pill treatment groups can downgrade the Survivin expression of cancer cells.Conclusion:Rhubarb zhi bug pill can restrain the tumor 's growth of S180 tumor-burdened mice, and promote the apoptosis of tumor cells in tumor-burdened mice, which point to Rhubarb zhi bug pill can be a effective antitumor drugs.
