Ojective: The objective of this study was to evaluate the possible role of IL? polymorphism (C590-T) in modulating asthma of children.Methods:The study was conducted in two different groups :asthmatic children(n=27) and healthy children (n= 30). The IL-4 C590-T polymorphism was genotyped by a polymerase chain reaction-restriction fragment length polymorphism analysis (PCR-RFLP). The serum 1gB levels were determined by the Enzyme-linked immune sorbent assay(ELISA).Ail data were made statistical analyses by using the FISH exact method, t test and the analysis of variance.Results : There was rio significant difference in the distributions of the genotypes(CC. CT and TT) between the2asthmatic group and the control group (P> 0.05) .Frequency of the T allele was 0. 648 in the asthmatic group and 0. 583 in the control group. The frequency of the T allele was not different between the two groups (P>0.05) .No significant difference i.n the levels of total serum JgE was observersed between subjects with a homozygote for the C allele (CC),a heterozygote (CT) and a homozygote for the T allele (TT) by the asthmatic group and the control group comparisons. There was no significant difference in high IgE level group compared with normal group too (P > 0. 05)Conclusion Our data suggest that the IL-4 promoter C590-T polymorphism is not associated with the development of asthma in children and does not play in modulating total serum 1gB levels. |
