The anti-inflammatory and immune action of 4igularia flscheri was studied in rats and mice. Ligularia fischeri (Lf) is the extract from the above ground part of Ligularia fischeri in ethyl alcohol. Ligularia fischeri (Ledeb.) Turcz., also named L.speciosa Fisch.et. Mey;L.sibirica Cass.Var.speciosa DC., is a perennial herb, belonging to Compasitae. Its root and rhizome, called Radix et. Rhizoma Ligularia, has long been used in the treatment of cough in Chinese traditional folk medicine. It is used to relieve cough and remove sputum. As to its pharmacological actions, Research has been limited to its expectorant and antitussive effect. A systematic report has not been seen on its above ground part or anti-inflammatory and immune actions. In our experiment, we report the theraputic effects of Ligularia fischeri on acute, chronic and immune inflammation in rats and mice by using several study models, and its mechanism is discussed preliminarily. The purpose is to provide a scientific basis for clinical application and further research. Methods and results are as follows:1.Effect on swelling of the earflaps of mice induced by xylene, croton oil.Dose:NS 20m1/kg, Lf2000, 1000, 500mg/kg, Indomethacin2Omglkg. After administration of the drugs, xylene or croton oil was applied to the, right earflaps and inflammation was induced. Weight difference between the same size of the two earflaps was indexed and degree of swelling and inhibition rates were calculated. The results showed that Ligularia fischeri significantly inhibited, in a dose-dependent manner, the swelling induced by xylene and croton oil in the ear of mice, and there was significant difference between Lf and lndomethacin.2.Effect on swelling of hind paws induced by injection of carrageenin or fresh egg white in rats. Dose: NS lOml/kg, Lf 1600,800,400mg/kg, md 20mg/kg.After administration of the drugs, carrageenin or fresh egg white was injected under the aponeurosis of the right hind plantar paws to induce inflammation. Each volume of the right hind paws was measured at different times and inhibition rates were calculated. The results showed that Ligularia fischeri dramatically inhibited the swelling of hind paws induced by carrageenin or fresh egg white in a dose-dependant manner and the effectivc time lasted for 6 or 4 hours. There was difference between Lf and lndomethacin.3.Effect on proliferation of granuloma induced by cotton-pellets in mice. Two aseptic cotton-pellets were implanted in both .subcutameous scapular regions. Dose: 20rnl/kg, Lf2000, 1000, 500mg/kg, md 20mg/kg. The drugs were administered daily for 7 days. On the 8th day, after being removed, the granuloma were dried and weighed, the weights of spleen, thymus and adrenal glands were also weighed. The results showed that Ligularia fischeri inhibited, in a dosedependent manner, the proliferation of granuloma, the effect accompanied by splenatrophy and megalothymus, while the weights of adrenal glands of rats were not affected. There was difference between Lf and lndomethacin.4.Effect on adjuvant arthritis in rats. Dose: NS 2OmlIkg, Lf1600, 800,400mg/kg, Fruends complete adjuvant was injected into the intracutaneous of the right hind paws to induce arthritis. The drugs were administrated daily for 3days, from the 8th day, the drugs were given daily for 7 days more. The volumes of the right hind paws were measured and inhibition rates were calculated at different time. The body reaction of rats was also observed. On the 24th day, the body, spleen, adrenal glands and thymus of the rats were weighed. The results showed that Ligularia tischeri had certain inhibitory effect upon adjuvant arthritis in a dose-dependent manner, and increased the weights of the body, the spleen, decreased the weights of thymus, while Ligularia fischeri did not affect the weights of the adrenal glands.5.Effect on scald edema in rats.
