Traditional Chinese drug is our traditional weapon of curing disease. It contains great vitality. The study of isolating natural drugs' components, purifying natural drugs, mechanism of action have developed many years. The medicinal chemistry has been the attentive hotspot of chemistry and biology. The saponin is an important bioactive substance, and it has remarkable resistance of bacterial activity. The steroid sapogenin is the chief raw material of composing steroid contraceptive and hormone. With the development of studying means in isolation technique and fabric research, the saponin chemistry is developing quickly. Recently, with the discovery of anticancer and bacteriostasic activity, et al., the study of steroid saponin developed quickly. The study has been taking important position in natural product chemistry. Systematically studying the chemical activity and biological activity of steroid saponin, searching the active lead compound, deeply developing new drugs, are always the people's studying goal. This paper studied the bacteriostasic activity of traditional Chinese drug and steroid saponin. The thermal power-time curve of bacterial normal metabolism and the bacteriostasic activity of saponin were determined by 2277 Thermal Activity Monitor. This paper analyzed the thermal power-time curve, constructed the theoretical model and thermodynamic equation of bacterium growth, and studied the thermodynamic characters. The study will provide theoretical basis for instructing to compose new antiseptic specific medicine. 1. The chief contents and conclusions1.1 The extraction of steroid saponin in yam, ophiopogon root, radix polygonati officinalis, smilax glabra, Allum macrostemon Bunge, rhizoma anemarrhenae.Most saponins can dissolve in water, ethanol, methanol, but can not dissolve in ether, trichloromethane, benzene, and other organic solvents. So we always extracted saponin using ethanol, methanol, and other polar solvents. Then we abstract it with petroleum ether, ether, de-fat, detach pigment, and other impurities. But the dissolvability of some steroid saponins in some aqueous alcohol is more than those in some pure alcohol. So we use alcohol with different purity when we extract them. For example, we use 80% ethanol to ophiopogon root, 95% ethanol to radix polygonati officinalis, 95% ethanol and 60% ethanol to smilax glabra.We can pure the steroid saponin when we extract them. The generalmethods are solvent extracted method, solvent deposited method, adsorption method with MgO, deposition method with cholesterol, deposition method with lead salt, chromatography, et al.. This experiment used solvent extracted method and solvent deposited method. We dissolved the impure saponin in water, and extracted it three times with equal volume water-saturated n-Butyl alcohol. Then we recovered the n-Butyl alcohol, dissolved the saponin thick paste in little alcohol, add acetone drop by drop several times to alcohol volume, shaken even as adding. At that moment, some saponin separated from them. We filtered them with Buchner funnel, and dried them with water extractor.1.2 The study of the rule that the E: coli grew and metabolized in traditional Chinese drug extracts and the steroid saponin solution with different concentration and the bacteriostasic effects of drug.For determining the rule of the drug function to E. coli and the optimum concentration of bacteriostasic activity, we add Chinese drug extracts with different concentration separately in liquid medium contained bacterium. The suspending liquid is formed. Then we measured the thermal output-time curve that the E. coli grew in 31 OK. By Logistic equation, we dealt with the exponential phase of growth curve, got the growth speed constant in Chinese drug extracts with different concentration, got the relation between the growth speed constant and concentration, determined the optimum growth concentration and the optimum bacteriostasic concentration. The results showed the traditional Chinese drug we choose could resist the growth of E.
