The root tuber of the Cynanchum auriculatum Royle ex Wight of the genus of Cynanchum in the family of Asclepladaceae is the main origin of the TCM drug Baishouwu. The constiruents and quality contral standard of the root tuber of the Cynanchum auriculatum are studied in this thesis.The thesis is divided into three chapters:Chapter 1 is a review of the studies on the chemical constituents of the plants belonging to the genus of Cynanchum at home and abroad from 1990 ; the herb classic textual research of Baishouwu; the research of its raw botany; the research of its preparation and its pharmacological effect.Chapter 2 discusses the extraction, separation, preparation and standardization of the standard substance of the C21 steroid glycosides in Baishouwu.C21 steroid glycosides are the characteristic and main effective constituents of Cynanchum auriculatum. They are also the symbol constituents to contoll the quanlity of the drug. The higy quantity constituents cynauricuoside A,cynauricuoside B, wilfoside C1G, wilfoside K1N and wilfoside C1N are selected as the control standard substances. Their extraction method and purification method are studied. High purity compounds are obtained by the positive phase columns chromatogram, reverse phase columns chromatogram and prepare HPLC column. Purity is analysised by the TLC and HPLC. Structure is identified by the UV spectrum, IR spectrum, MS spectrum,1H-NMRspectrum and 13C-NMR spectrum. Some standard substances are obtained. The stuadies in this chapter provided the stable and credible method to separate and prepare the C21 steroid glycosides standard sustance. It will also benefit the studies on the medicinal material quality control standard. Chapter 3 discusses the sutudies on the Baishouwu medicinal material quality control standard.There is no report about the studies on the Baishouwu medicinal material quality contral standard. Two novel methods for the quantitative determination of C21 steroid glycosides in Baishouwu, colorimetric method with vanillin-vitriol solution as the reagent and RP-HPLC method, are established. The colorimetric method used cynauriculoside A as the standard substance to detect the toal glycosides. The HPLC method can detect 4 compounds at the same time, λ=220nm, and the isacratic solution as the flow phase. The methods can be used as an objective standard to estimate the quantituy of the medicinal material and its preparation. Quantitative determination of C21 steroid glycosides in Baishouwu which are prepared in different ways or collected in differrent months is completed. Change caused by the preparation process to C21 steroid glycosides is discussed. Among the samples collected in different months, the total glycosides is maximum in the sample collected in October. The result is identical with the reports in herb classic. The results show that the heat can reduce the tatol glycosides in Baishouwu. It can be conclused that in the prescriptions and preparations in which the C21 steroid glycosides are the main effective constituents the raw medicinal material is better. The ratio between the each compounds changed in the prepare process. It can be conclused that cynauriculoside A and wilfoside C1G hydrolyzed and lost 1 molecule glucose, transformed into wilfoside K1N and wilfoside C1N in the process. Based on these work, a quantity criterion draft about Baishouwu is drawn out. It provided the standard to controll the quality of Baishouwu medicinal material. Summarize the article, five C21 steroid glycosides standard sustance are obtained from the medicinal material, that is, cynauricuoside A,cynauricuoside B, wilfoside C1G, wilfoside K1N and wilfoside C1N. The studies on their standardization is carried out for the first time.Two new methods, colorimetric and RP-HPLC, for the quantitative determintion are established for the first time. To control the quality, a quantity criterion draft about Baishouwu is drawn out.
