Nitric oxide (NO) is one of the focus of biomedical study in recent years. NO,a comman gas matter with simple structure and instability , is generated by the catalysis of nitric oxide synthase ( NO syn-thase)in human body. As vasculodilator and neurotransmitter, different typed NO synthase exert different roles at differential phases .In recent years the experiment of domestic and international bulk analysis animal have already definited the generating route and physiology effect of NO, as well as the action mechanism in the damage of cerebral ischemia.Many researchers studied the variety of NO contents in the plasma of cerebral infarcton patient, and obtained consistent conclusions; but the reserachs for the patients of forebrain arteriosclerosis and transient ischemic attack ( TIA) are not much , and the conclusion is also not the same.Referencing the former bulk data of the animal and the clinical experiment , our study furtherly expatiate action mechanism of NO at cerebral ischemia , provide the experiment for applging NOS inhibitor at the best time , and look fov the new treatment trial for the cerebral ischemia patients.MATERIALS AND METHODSControl group: 40 instances, cerebral infarction group: 40 instances, TIA :20 instances, cerebral arteriosclerosis 20 instance.Adoptting the morning cubital venous blood 2.0 milli liter of suffers, place it aside fov 2 hours, then centrifugalizing 10 minutes by 3000rings per minute at room temperatrue. take the plasma in theplastics test tube, the latter is seal and taken into a refrigerator( -20. C ) standby.NO examination adopts nitrate reductase method and the reagents are offered by Nanking Jiancheng Biological engineering research insi-tute.EXPERIMENT RESULTS1. The NO contents in the cerebral infraction group plsma increase prominenly in acute phase ( P <0.01) and have none statistical significance in resume phase( P > 0.05).2. The NO contents in the TIA group plasma increase prominently ( P<0.01) .3. The NO contents in the cerebral arteriosclerosis group plasma have none statistical significance ( P>0.05) .DISCUSSION1. The NO contents variety of cerebral infarction plasma In recent years many domestic and international animal experiment express that the NO have nerve perfection and toxicity actions -two different actions in the cerebral ischemia or ischemic reperfusion damage . About cellular toxic action of NO, past researches have already expressed, besides lie the nerve toxicity action of the glutamic acid, high - concentration NOS inhibit various enzymes relating to e-lectric charge transmission system and citric acid cycle of mitocondri-ons , common catalytic actives center--nonheme iron - sulfur complexes, at the end inhibite mitochondrion respiratory chain. On the other hand, the interaction of NO and super - oxidase anion( 02 - ) may formulate peroxide nitroso anion( CNOO - ) , the latter decom-pose the strong toxic oxindole ( OH - ) and nitrogen dioxide ( N02 - ) free radical. Experiment results indicate that NO contents in plasma of cerebral infarction change , expressing the NO impacting cerebral infarction.2. The variety of NO contents of TIA plasmaThe plasma NO contents of TIA group increase prominently. Its mechnism is that the synthetase and releasing of NO in the endothelial cells of vessels decrease , which reduce the reactivity of abreent vessels to vasoconstrictors higher, causing the spasma constriction of vessels with heavier pathologic changes. As the pathological changes blood vessel , the supplying blood flow of decreases sharply , appearing sluggish flow. In the vessels with lighter pathological changes , endothelium accelerate the synthesis of NO and other vasodilators and platelet aggregations compensatively , to improve blood circulation. When compensation is strong , there will be a transient cerebral ische-mic attack.3.
