Objective To investigate the expression of tumor suppressor gene PTEN/MMAQ /TEP, (short for PTEN as follow) protein and integrin α5β1 and the role of them in malignant biological behaviors and angiogenesis in gastric cancer(GC)Methods Immunohistochemical staining (S-P Method) was employed to detect the expression of PTEN protein and integrin α5β1 in 35 primary gastric carcinoma (PGC) , 10 lymth node with metastasis and 8 chronic superficial gastritis (CSG) and CD34 expression for microvessel density (MVD) in 35PGC and 8 CSG on paraffin-embeded sectionsResults (1) Compared with the CSG group, a strinking expression decrease of PTEN protein (t=2.45, P<0.05) and a marked overexpression of integrin α5β1 (t=3.32, P<0.01) in PGC group were showed respectively, however ,in preinvasive GC, No significant expression changes were observed between the expression ofPTEN protein (t=0.46, P>.0.05) and integrin α5β1 (t=0.15, P>0.05). Compared with those in CSG while in advanced PGC, there are notable expression change of them as compared with those in GC in situ (t=1.85, 1.90, P<0.05) respectively (2) the inactivatien of PTEN in PGC shows a notable association with both lymph node metastasis (t=2.92, P<0.01) and age (t=2.26, P<0.05) while overexpression of integrin α5 β1 is only associated with the invasion depth( t=2.29, P<0.05); In metastasic lymph node carcinoma, the expression of PTEN shows a slight decrease (t=0.73, P>0.05) while that of integrin α5 β1 shows a striking increase (t=2.45, P<0.05) as compared with those in corresponding PGC respectively. (3) MVD shows a significant association with all the invasion depth (t=3.37, P<0.01) , lymph node metastasis (t=2.49, P<0.05) and TNM stage (t=2.94, P<0.01); In PGC in situ, MVD is similar to that in CSG (t=0.26, P>0.05) and sigmificantly lower than that in advanced PGC (t=2.31, P<0.05); MVD in PTEN-negative PGC is much more higher than that in PTEN-positive PGC (t=2.60, P<0.05) and a significant inverse correlation is shown between the expression of PTEN protein and MVD (γ= -0.363, P<0.05), however, Not a correlation of expression of integrin α5β1 with MVD ( γ=0.156, P>0.05). (4)the expression of PTEN is not correlated with that of intgrin α5 β 1 (t=0.22, P>0.05). In PGC with PTEN(-)/integin α5β1 (+), the rates of all the scirrhous invasion, lymph node metastasis , TNM III/IV stage and high MVD were higher thqn those in the other PGC respectively (P=0.002, 0.07,0.041,0.041).Conclusions Our results suggest that the decreased expression of PTEN protein and the overexpression of integrin α 5 β 1 and MVD are all demonstrated in PGC, especially in advanced PGC, The expression status of them paly a key role in the influence on biological behaviors and the evaluation of prognosis of GC; PGC with PTEN(-)/ integrin α 5 β 1 (+) might have a worse outcome for it's much more rich in the potential of invasion and metastasis; The inactivation of PTEN gene is in close correlation with the angiogenesis activity of GC.
