Pain Cream Anti-inflammatory And Analgesic Effects Of Transdermal Drug Delivery Safety Of Experimental Research

Objectives:Studies on the Effects of Anti-inflammatory and Analgesic of Chinese traditional medicine pharmaceutics of GU TONG CREAM,and the reliability of skin external use of GU TONG CREAM.Making the foundation of discovering the mechanism of GU TONG CREAM.And so provide a theoretic basis for the clinical application of GU TONG CREAM.Methods:1 Anti-inflammatory test:1. 1 Foot swelling:40 male rats,divided equally into 4 groups: group of base cream,group of GU TONG CREAM I , group of GU TONG CREAM II ,and group of TIAN HE GU TONG cream.Administration on the first,third,fifth,and seventh day.2 hours after the last administration, inject 0.1ml 10% fresh egg schedule subcutaneously in the middle of rat's left hindfoot.After injecting for 0.5,1,2,4,and 6 hour,assay the volum of the rat's right and left hindfoots.Indicate rat paw edema by difference of the rat's right and left hindfoots.1. 2 Auricle swelling:40 male mice,and the group dividation and administration are same as experiment 1.1. 2 hours after the last administration, Inunct 0.05ml dimethylbenzene on the right auricle.20 minutes after killing the mice,cut circle auricle of every mice's left and right auricle.The difference of the circle auricle is the edema of the auricle.2 Analgesia test:2. 1 Hot plate test:Assay the female mice's background pain threshold on the 55±0.5 ℃ hot plate.Choose 40 mice according with standard,and the group dividation and administration are same as experiment above. 2 hours after the last administration,assay the mice's pain threshold again.2. 2 Writhing test:Choose 40 male mice,and the group dividation and administration are same as experiment above. 2 hours after the last administration,inject 0.2ml 0.6% glacial acetic acid into mice's abdominal cavity.Record the times of wrenching their body in 15 minutes.3 Acute toxicity:60 white cavy,divided equally into 6 group:group of base cream(integrity skin),group of low dosage GU TONG CREAM(integrity skin), group of high dosage GU TONG CREAM(integrity skin), base cream(damaged skin),group of low dosage GU TONG CREAM(damaged skin),and group of high dosage GU TONG CREAM(damaged skin).After using narcotics,observe weigh,skin, mucosa,breath and activity.Result:1 Anti-inflammatory test:GU TONG CREAM significantly inhibit rat paw edema and dimethylbenzene-induced auricle edema.2 Analgesia test: GU TONG CREAM increases mice's pain threshold and reduces the times of writhes induced by acetic acid.3 Acute toxicity:The groups of GU TONG CREAM had no acute toxic reaction and no death.ConclusionGU TONG CREAM has significant analgetic effects and notable effects of anti-inflammation.And it is safe for dermatologic.
