Objective:Heilongjiang Province is rich in Geranium herb resources, which has a variety of pharmacological effects, such as anti-inflammatory, analgesic and antibacterial, antiviral, antioxidant, hypoglycemic, protecting liver and preventing cough role. Study of Geranium herb about the chemical composition of its active substances and the relationship between drug action is of great significance to its full excavation, the use of Geranium resources, as well as the research of new drugs, which will accelerate the Northern drug development and application.Methods:1, Focused on(Geranium wilfordii Maxim.) by using water extraction, ethanol extraction, and acetone extraction methods to extract its effective ingredients. Through the animal experiment, the findings showed that the extraction had better anti-inflammatory,analgesic and anti-diarrhea effect to the mice.2, The better extraction was successiveness extracted with petroleum ether, ethyl acetate, n-butanol extraction, the experimental results were obtained by the product acting on the above mentioned mouse models to affirmed the best part for the secondary extraction components.3, By the way of Silica Gel Column Chromatography, ODS Reverse Phase Column Chromatography, RP-HPLC and NMR spectral et al to analyzed the best part.Results:1, In the inflammatory model, water extraction group(t=4.33,P<0.01),ethanol extraction group(t=5.07,P<0.01),acetone extraction group(t=5.81, P<0.01);in the analgesic model, water extraction group(t=5.10,P<0.01),ethanol extraction group(t=5.54, P<0.01),acetone extraction group(t=6.10,P<0.01);in the anti-diarrhea model, water extraction group(t=6.06, P<0.01),ethanol extraction group(t=8.40, P<0.01),acetone extraction group(t =8.55,P<0.01).2, In the inflammatory model, the ethyl acetate part of the high dose group (t= 9.42, P<0.01), ethyl acetate part of the middle dose group (t=2.44, P<0.05), aqueous part of the high dose group (t=8.58, P<0.01), they were significantly different compared to the model group, with obvious anti-inflammatory effects; in the analgesic model, ethyl acetate and some of the high dose group (t=10.58, P<0.01), ethyl acetate part of the middle dose group (t=2.22, P<0.05), n-butyl alcohol high dose group (t=8.56, P<0.01), aqueous part of the high dose group (t=8.59, P<0.01), they were significantly different compared to the model group, there are obvious analgesia; in the anti-diarrhea model, ethyl acetate, the high dose group (t=8.85, P<0.01), ethyl acetate dose group (t=2.47, P<0.05), n-butyl alcohol high dose group (t=7.82, P<0.01) and aqueous of the high dose group (t=7.62, P<0.01), they were significantly different compared to the model group, with an obvious anti-diarrhea effect.3, It was isolated five compounds from the acetone extract of ethyl acetate extract of Geranium wilfordii Maxim. Conclusion:1, Using water extraction, ethanol extraction and acetone extraction methods to extract Geranium wilfordii Maxim. Actone extraction had best drug action.2, The comprehensive analysis affirmed that the ethyl acetate fractionation is best part for the secondary extraction components.3, In the best active extract, it includesβ-sitosterol, Quercetin, Kaempferol-7-O-rhamnoside, Kaempferol, and Gallic Acid.4, In the safety tests for mice, the mice were fed with Geranium water extract, ethanol extract and acetone extract, and there was no manifestation of toxic, suggesting that the extract of Geranium wilfordii Maxim, with effective therapeutic dose is actually safe.
