Objective:This study was designed to establish a hepatitis B virus infection acquired(DHBV) model ferruginea,Yin Shao Powder casual observation in vivo the role of anti-DHBV DNA, preliminary study of its mechanism of action.Methods:(1) model:The PCR method DHBV DNA positive serum screening,with positive serum DHBV DNA by duck duck leg venous inoculation,the day after the establishment of animal models infected with DHBV duck.(2) Experimental observation:Duck hepatitis B animal models 40 were randomly divided into 5 groups:model control group,Yin Shao Powder scattered high,medium and low dose group and lamivudine group.Model control group given conventional feed,free drinking water;Yin Shao Powder casual drug use high-dose group 40g.Kg-1.d-1,in drug dosage group 20g.Kg-1.d-1,low-dose drug group 10g. Kg-1.d-1,lamivudine group medication 50mg.Kg-1.d-1,a one-time oral administration.Before treatment,drug intervention in the first days 7,14,21 and 7 days after stopping taking the first leg vein 0.5-1ml,3000r/min centrifugal 10min,serum prepared seized -20℃.Pre-treatment DHBV-positive ducklings sacrificed 2,drug intervention in the first 21 days post-kill Yin Shao Powder scattered high-dose group and lamivudine group 2 of the ducklings,respectively, from liver tissue fixed 4%formaldehyde solution prepared seized.(3) laboratory tests:using fluorescence quantitative polymerase chain reaction(FQ-PCR) Detection of serum DHBV prepared seized content,liver tissue HE staining under light microscope(×100) to observe the pathological changes in liver at the same time testing the serum of each group C acid aminotransferase(ALT) and aspartate aminotransferase(AST) levels.Results:(1) lamivudine group 7 days after administration of serum DHBV DNA levels rapidly decreased during the treatment has been maintained at a relatively low level of serum 7 days after stopping the virus titer rise,when compared with the model control group no significant difference(P>0.05).Yin Shao Powder scattered high,medium-dose group 14 days in the administration of serum DHBV DNA content began to decrease significantly decreased in 21 days,with the same point in time the model control group were significantly different(P<0.05),with Pascal Lamy at the same time stavudine group no significant difference(P>0.05); drug 7 days after the Yin Shao Powder high,medium-dose group still demonstrated a certain degree of virus inhibition,with the same point in time lamivudine group were significantly different(P<0.05).Yin Shao Powder scattered low-dose group during the treatment no significant inhibition of the virus.(2) Yin Shao Powder high dose group were lower in ALT,AST levels than lamivudine group at the same time the difference was significant(P<0.05).(3) Yin Shao Powder of liver pathology showed that high-dose group scattered to reduce the effect of liver inflammation than lamivudine significantly.Conclusion:Yin Shao Powder scattered DHBV could inhibit the replication and reduce liver inflammation,and its efficacy and dose,treatment time is related to the role of its anti-DHBV more durable than lamivudine.
