Objective: In order to set up a post-stroke depression rat model. To investigate the influence of YuLeShu on the depressive behavior and cerebral monoamine neurotransmitters of the PSD rat model. Furthermore, to probe the anti-depressive mechanism of YuLeShu.Methods: Set up a focal cerebral ischemic rat model by occluded right side middle cerebral artery. On the basis, further isolated-living condition and moderate unpredictable stress are applied successively to set up PSD rat model. The neurological functional recovery of stroke rats is assessed by Longa 5 grades. Sucrose solution consumption test and open-field test are used to estimate the change of interest, the level of spontaneous motor activities and investigative behavior of PSD rat model. Fluorophotometry is applied to detect the monoamine neurotransmitters(NE 5-HT and DA) of the double frontal-parietal cortexes and brain stem. Evaluating the efficiency of YuLeShu on the depressive behavioral tests and cerebral monoamine neurotransmitters of PSD rat model.Results: The PSD rat model manifests significant changes: neurological functional impairment, slow growing, the decrease of sucrose solution consumption, the dropping of open-field behaviors, the dropping of cerebral monoamine neurotransmitters. YuLeShu can augments the consumption of sucrose solution, improves the open-field behaviors and increase cerebral NE and 5-HT concentration of PSD rats significantly.Conclusion: On the basis of MCAO rat model, isolated-living condition and moderate unpredictable stress are applied successively to set up suitable PSD rat model. YuLeShu can ameliorate the depressive behaviors and cerebral monoamine neurotransmitters concentration of PSD rat model. The anti-depressive mechanism of YuLeShu manily is due to increases the cerebral monoamine neurotransmitters concentration and the systemic promotion cerebral function.
