Objective To investigate the co-function of HBx gene and IGF-II gene in the pathogenesis of hepatocellular carcinoma(HCC) and elucidate the molecular mechanism of HCC carcinogenesis. Methods Semi-quantitive reverse transcriptase-polymerase chain reaction (RT-PCR) was adopted to detect the expression variation of HBx gene and IGF- II gene in 30 HCC tissues, their corresponding adjacent liver tissues and in normal liver tissues of 10 non-liver disease patients. Results (1)The positive expression rate of HBx in 30 HCC tissues,their corresponding adjacent liver tissues and 10 normal liver tissues of non-liver disease patients was 86.7% (26/30) ,1% (3/30) and 0% (0/10) respectively;the expression quantity of HBx in HCC tissues and the adjacent liver tissues was 0.752 0.212(x s) and 0.010 0.002( x s) respectively. There was significant difference between them(P<0.001);(2)The expression quantity of IGF-IlExon4 in the HCC tissues,its adjacent tissues and normal liver tissues was 1.193 1.063 (x s), 0.043 0.023 (x s) and 0.011 0.005 (x s) respectively. The expression quantity of IGF- IIExon4 gene in the three tissues was significantly different(P <0.001),however no significant difference between the HCC tissues and its adjacent tissues; (3) The expression quantity of IGF-II Exon4B in the HCC tissues and its adjacent tissues was 0.103 0.020 (x is)-, 0.101 0.017 (x s) respectively. The different between the two tissues was no significance(P>0.05);(4)The expression quantity of IGF-II Exon4 was 0.474 0.143(;c s)-, 0.016 0.002 (x s) respectively in HCC tissues and its adjacent liver tissues which express HBx and IGF- II Exon4 simultaneously.The expression quantity was significant different(P<0.01);(5)The expression quantity of IGF-IlExon4B was 0.507 0.147(x s)., 0.035 0.006(;c s) respectively in HCC tissues and its adjacent liver tissues which express HBx and IGF- II Exon4 simultaneously.The expression quantity was significant different(P<0.05); Conclusions (1) The expression quantity of HBx in HCC tissues was high;(2) The expression quantity of IGF-II gene in HCC tissues and its adjacent tissues was greatly higher than that of the normal tissues, which indicated that the overexpression of IGF-II gene was closely related with the carcinogenesis of HCC; (3) HBx may trans-activate the transcription of IGF- II gene; (4) As the potential tumor markers,HBx and IGF- II gene may own great value for the early gene diagnosis of HCC.
