Metallothioneins(MTs) are a family of low molecular weight, cysteine-rich, metal-binding proteins, widely distributed in nature and generally considered responsible for heavy metal detoxification, free radical clearance, against ionizaion radiation and essential metal metabolism in eukaryotes .In this paper, we used an effective MT inducer-organic seleniumm which has been confirmed by my parter to induce the biosynthesis MT in rat liver. Isolation, purification and identification of metallothionein in Wister rat liver by Gel filtration, SDS-PAGE. According to the propery of MT , we attempt to isolate, purify and identify MT by protein finger mapping. To research the MT protective effection against pulmonary injury induced by ionizaion or radiation.The metallothioneins purified by classical method have been confirmed consistent with Sigma MT standard. Analysis the elements of MT indicate sulfur content of control group is higher about two folds than that of selenium group, whereas selenium content is lower about two folds. So we confirm primarily that the sulfur atom located in cysteine of metallothionein was replaced by selenium atom and may be find a novel MT. The result from pathology and biochemistry indicated selenium group has the protective effect against lung injury induced by radiation. By serum protein fingerprint we also confirm MTs have the protective function against radiation and for the first time we identified the MT from lung tissue by the SELDI-TOF-MS.
