To investigate the possibility of the association of ARX gene with children's intelligence in Qin-Ba mountain area, studies were carried out among children from two different areas Zha Shui and An Kang two experiment centers in Qin-Ba mountain area. There were 550 children involved, in which 116 children were of low intelligence, 114 children were of marginal intelligence, and the other 320 children were of normal intelligence. The Microsatellite DNA DXS8027 and DXS8056 that tightly link with ARX gene were used as genetic marker to investigate the allele distributions and make association analysis with intelligence character. After analysis with various methods, such as PCR, PAGE, T-cloning and DNA sequencing, the DXS8027 allele, but not DXS8056, was chosen to be used as the molecular weight marker. The results showed that there were significant differences in the distribution of DXS8027 allele frequence between male and female (X 2=30. 242, p<0. 01) , but the differences was not significant between the same sex groups of the two area(X 2=4. 703, p>0. 05, X 2=14. 952, p>0. 05) , or between the two areas regardless of sexes (X 2=15. 2, p>0. 05) . The distribution of DXS8027 allele frequence in all 550 children was in accordance with Hardy-Weinberg balance, and the heterozygosity(Het=0. 7968) was higher than European (Het=0. 6630) , Compared with the previous study we can get the more valuable polymorphism information. Chi-squre test was used to compare the allele distribution between low intelligence group, marginal intelligence group and normal intelligence group. All the results showed that the DXS8027 A5 is associated with normal intelligence and DXS8027 A7 is associated with low intelligence (p<0. 01). The results suggested that: (1) There may exist different ARX allele in ARX loci exerting different effects on intelligence development; (2) DXS8027 A7 allele may have linkage with ARX gene with harmful mutation.
