Objective To find out the basic Traditional Chinese Medicine pathologensis of chronic fatigue syndrome, and create chronic fatigue syndrome animal model.To study the mechanism of chronic fatigue syndrome in molecul and rudimen clarify the mechanism of anti- fatigue prescription therapying chronic fatigue syndrome.Method (1)Wistar male rats were divided into 3 groups :normal group,model group, anti- fatigue prescription group.Using compound agents to make chronic fatigue animal model:rats accepted 14 days compound agents' stimulation,that is to say ,on odd-numbered days swims in cold water for 30 minutes, on even-numbereddays mated with rut female rats; Every day clips the rat's tail to stimulate for 3 hours. (2)in open field,strength exhaust,swim and rat's tail suspending to observe the change of praxiology.(3) Using radio-immunity to detect the content of immunoglobulin A , immunoglobulin G and immunoglobulin M in blood serum.(4)Detecting the content of 5-HT, dopamine and adnephrin in plasm using radio-immunity.(5)Detecting the change of thromboxane B2 and 6- keton-prostaglandin in plasm using radio-immunity. (6) Taking transmission electron microscope to detect rat's adrenal cortex ultrastructural. (7) Using radio-immunity to detect the content of monoamine inHypothalamus and pituitary gland.Result (1)The model group has phanero-anomaly changes: body weight descent, hair abscission, lazy, stool no- shape, appetite descent,and easy to rage,et al.(2)The content of immunoglobulin A , immunoglobulin G and immunoglobulin M in blood serum was lower than the nomal, anti- fatigue prescription group's change significant relieve.(3)The content of 5-HT, dopamine and adnephrin in plasm was higner than the nomal, anti- fatigue prescription group's significant descent. (4) The content of thromboxane B2 and 6- keton-prostaglandin in plasm was higher than the nomal, anti-fatigue prescription group's near to the nomal.(5)Adrenal cortex ultrastructural: the model group's lipid vacuoles was less than the nomal, mitochondrion blanket was rarefaction and swelling, anti- fatigue prescription group's near to the nomal.(6)The content of monoamine in Hypothalamus and pituitary gland of model group were lower than the nomal, anti- fatigue prescription group has no difference with the nomal.Conclusion We succeeded in creating the chronic fatigue syndrome animal model,and suggested standpoint that " excess of liver,deficiency of Spleen and Kidney" is the basic Traditional Chinese Medicine pathologensis of chronic fatigue syndrome.We found that model rats' behavior changed , the content of immunoglobulin A , immunoglobulin G and immunoglobulin M in blood serum was low, thromboxane B2,6- keton-prostaglandin F1a,5-HT,dopamine and adnephrin washigh, the content of monoamine in Hypothalamus and pituitary gland of model group were lower than the nomal, it makes clear that the model rat was fatigue, various kinds of compound stress result in correlated biochemistry factors' disorder was the main mechanism.The study manifest that anti- fatigue prescription can adjust and recovery the factors,we suspect it can regulate the nerve-endocrine-immunological network.The anti- fatigue prescription's effect disproof that chronic fatigue syndrome Traditional Chinese Medicine pathologensis is reasonable and scientific.
