Objactive:To study the expression and significance of Caspase-3, Survivin, CyclinD1 and P27 in gastric carcinoma(GC). To investigate the correlation of proteins referred to above and the relationship between the biomarkers and clinical pathological parameters. To analyze the significance of the biomarkers in the occurrence and development of GC, to study the significance in diagnosis, therapy and prognosis of GC so as to provide the valuable evidence for the early clinical diagnosis and genetic therapy of GC.Method:Non-Biotin HRP Detection system immunohistochemical Ready-to-use two-steps method was performed to research the expression of Caspase-3,Survivin,CyclinD1 and P27 proteins in 72 cases of GC, 9 cases of dysplasia and 54 cases of normal gastric mucosa. Results were assessed by SPSS version 12.5.Results:1. There was obvious difference about Caspase-3 protein expression in GC, dysplasia and normal gastric mucosa (P<0.01). The positive rates of Caspase-3 protein expression were significantly higher in the cases of better histological differentiation than the cases of worse histological differentiation and in the cases of clinical stage I ~ II than the cases of clinical stage III~IV (P<0.05).2. The difference of Survivin protein expression was significant among GC, dysplasia and normal gastric mucosa (P<0.01). The positive expression rate of Survivin protein was related to clinical-pathologic characteristics of the GC (including lymph node metastasis, distant metastasis, clinical stages, Bormann type and Lauren classification).3. The expression level of CyclinD1 protein in GC was significantly higher than that in dysplasia and normal gastric mucosa (P<0.01). The higher expression of CyclinD1 protein was significantly correlated with worse histological differentiation. CyclinD1 expression had relationship with Bormann type and Lauren classification.4. The positive rate of P27 protein expression in GC was significantly lower than that in normal gastric mucosa (P<0.05). The abnormal expression of P27 protein was correlated with histological differentiation, depth of invasion, lymph node metastasis, distant metastasis and clinical stages of GC, for it was lower in GC with serosal infiltration, lymph node metastasis, distant metastasis, or late clinical stages than those without.5.
