Objective To explore the association between the polymorphism of metabolic genes(GSTT1,GSTM1,CYP1A1) and tumor suppression gene p53 coden 72 and human longevity.Methods The GSTT1,GSTM1,CYP1A1 genes and p53 codon 72 were genotyped with PCR-RFLP technique for 156 Zhuang healthily long-living people (aged 90-105,mean 93.2yrs,longevous group,LG) and 142 healthy controls (20-72,mean 34.8yrs, control group,CG) from Bama area,then the polymorphic combined genotype frequencies were analysed between the two groups. Results No significant difference was found in the frequencies of GSTT1(present or deleted), GSTM1(present or deleted), CYP1A1 (wild type T/T,heterozygous mutation T/C,homozygous mutation C/C ) between logevous and control groups.While the wild type genotype(G/G) frequency of LG was higher than that of CG(53%:29%,p<0.05). Analyses of various combining genotypes showed that, any simultaneous deletion or mutation of GSTT1,GSTM 1,CYP1A1 genes and p53 codon 72 was found more frequent in CG than LG. In particular, the combination of GSTT1(+)-GSTM1(-)-CYP1A1(T/C,C/C)-p53 coden72 (G/C,C/C) genotypes in male and female controls and GSTT1(-)-GSTM1 (-)-CYP1A1 (T/C,C/C)-p53coden72 (G/C,C/C) in female controls were markedly higher than accordingly longevous group.While the frequency of combined genotype of GSTT1(-)-GSTM1(-)-CYP1A1 (T/C,C/C)-p53coden72 (G/G) in male long-living subjects is greatly higher than male controls(p<0.05). Conclusions The (combining) polymorphism of metabolic gens GSTT1, GSTM1,CYP1A1 and tumor suppression gene p53 codon 72 might involve in the human longevity in Bama area which mechanism needs further studies.
