Si-Wu-Tang, a famous classical Chinese formula for treatment of blood deficiency, has pronounced curative effect on enriching blood, activating blood, regulating menstruation. Up to now, the molecular mechanism of Si-Wu-Tang for blood enriching effect has not been well documented yet. In order to clarify the blood enriching mechanism of Si-Wu-Tang at molecule level, changes of protein expression in bone marrow of blood deficient mice induced by CTX (cyclophosphamide) were studied using proteomics technique, on the basic study of blood deficiency induced by radiation. Changes of different expression proteins were confirmed by RT-PCR at mRNA level. The effect of Si-Wu-Tang to resist apoptosis of bone marrow cell induced by CTX was also studied. In addition, the effects of Si-Wu-Tang on proteins expression between two blood deficient mice induced by radiation or by CTX were rationally compared.It is shown that, expression of 12 proteins in bone marrow of blood deficient mice induced by CTX were up-regulated and 3 proteins down-regulated, respectively. These proteins involves GDP-dissociation inhibitor, cofilin, capping protein alpha 1 and visinin-like protein 1 which participate in cell migration and adhesion. Annexin A1 and PrX-V are associated with cell apoptosis.Carbonic anhydrase I and myeloid secondary granule protein involves in cell differention. The changes of mRNA of cofilin, annexin Al, PrX-V and carbonic anhydrase I are similar to their protein level. DNA Ladder results show Si-Wu-Tang has preventive effects by decreasing apoptosis of marrow cells after treatment with CTX for 6 h, and obviously attenuate the activity of caspase-3, but it has no effect on the inhibition of the up- regulated of mRNA of Bax. Using proteomic technique, by comparison of change in proteins between two blood deficient mice induced by radiation or by CTX, it is found that irradiation of 3.5 Gy 60Co γ will destruct the molecular network of bone marrow cells, and restrain the signal transduction of haematopoietic growth factor, inhibit genetic transcription and energy metabolism, block cell cycle, thus restraining differentiation of haematopoietic growth and proliferation.
