Objective : To study the effect of puerarin on chronic obstructive pulmonary disease rats.Methods : Sixty spragu-dawley rats were randomly divided into six groups: control group, COPD group, COPD+ Qi deficiency syndrome + puerarin group, COPD+ Yin deficiency syndrome + puerarin group, COPD+ blood stasis syndrome + puerarin group. The levels of tumor necrosis factor-α and ET-1 in serum were measured with ELISA. The level of IL-8 in BALF was measured with ELISA. The activity of SOD in bronchial and lung homogenates was determined by RIA method. Lung tissue section stained by HE was observed to study the morphological alternations, inflammatory cells of the bronchial wall were counted. Thickness of the bronchial smooth muscle layer and the pulmonary arterial wall were measured by image analyzer.Results(1) The level of the COPD model group's TNF-a(99.2±15.3 pg/ml), ET-1(9. 16 ±1.93pg/ml) and IL-8(115.4 ±23.8 pg/ml) were higher than the control group's TNF-a(56.7 ± 8.3 pg/ml), ET-1(3.66± 1.35 pg/ml) and IL-8(41.2± 11.0 pg/ml), P<0.01. The activity of SOD in the COPD model group (102.58± 15.38ku/gpro.) was significantly decreased compared to the control group (196.19 ± 8.86ku/gpro.), P<0.01. The numbers of inflammatory cells in the COPD model group was significantly increased compared to the control group (PO.01). The COPD model group's thickness of the bronchial smooth muscle layer(25.76±1.38 μm) and the pulmonary arterial wall(88.93±6.63 μ m) were significantly increased compared to the control group's thickness of the bronchial smooth muscle layer(17.27±2.28 μ m) and the pulmonary arterial wall(59.00± 11.16 μ m) (P<0.01).(2) The levels of the puerarin groups' TNF-a, ET-1 and IL-8 were lower than the COPD model group (PO.05).The activity of SOD in the puerarin groups were significantly increased compared to the model group (P<0.05).
