The crude polysaccharide was extracted from Sausurea involucrate Kar.et Kir. with hot water, and precipitated by ethanol. It was isolated and purified and structure of refined polysaccharide has been analyzed. Activity experiment of Sausurea involucrate Kar.et Kir. polysaccharide in scavenging superoxideradical hydroxylradical and lipid radical has been studied to provide scientific cue for the use of Sausurea involucrate Kar.et Kir..The crude polysaccharide XJXL has been analyzed by PC and GC, it was composed of Xyl, Ara, Rha, Gal, Glc, Gal-A and its molar ratio is 1.0: 7.0:1.7:2.3:1.7: 32.6.The crude polysaccharide XJXL has been eliminated protein and fractionated by acidic ethanol. Three fractions XL1, XL2, XL3 were got respectively. XL3 was purified by DEAE-Sephadex A-25 gel filtration, so XL31 was got. The identification of purification by Sepharose CL-4B, paper chromatography and cellulous acetate electrophoresis shows they were homogeneous. Their average molecular weight is 170 RD.PC GC analysis indicated that XL31 was composed of Ara, Rha, Xyl, Gal, Glc, GalA and its molar ratio is 11: 4:1:18:3: 9 Through reacting to pectins enzyme,IR, GC, periodate oxidation, Smithdegradation, methylation and GC-MS analysis, they reveal that XL31 is a kindof branched structure. The main chain is mainly maded up of β- (1â†'4) or β-(1â†'5) Ara, β- (1â†'6) Gal, β (1â†'4) Gal and a- (1â†'4) GalA. β- (1â†'4)or β- (1â†'5) Ara are substituted at 3-O and on average there are 2 branches among 9 main chain residues; (1â†'6) Gal are substituted at 3-O and 4-0 and on average there are 8 branches among 9 main chain residues; (1â†'4) Gal are substituted at 2-O , 3-O and 6-O and on average there is 1 branche among 1 main chain residues; (1â†'4) GalA are main chain residues and no branche; The side chain is composed of (1â†'4) , (1â†'5) -Xyl and (1â†'4) -Glc and (1â†'2, 4)-Rha. The nonreduced end is composed of GalA, Gal, Ara, Xyl, Rha, Glc.Activity experiment shows that Sausurea involucrate Kar.et Kir. polysaccharide have scavenging effect on ·OH- and O2-· but have no effect on R. Sausurea involucrate Kar.et Kir. polysaccharide have considerable antibiotic activity against bacteria, have no inhibition effects on epiphyte.
