Objective:To investigate the Isolation,purification,structural elucidation and antidiabetic activity of Polysaccharides from Alhagi-honey.Methods:1.Water extracting-alcohol precipitating method is applied to extract crude polysaccharide, AB-8macroporous resint,EDAE SepharoseCL-6B and Sephadex G-100 gel filtration chromatography is applied to isolation and purify of the polysaccharide.2.The purity and molecular weight of polysaccharide are determined by high performance liquid chromatography and qualitative of polysaccharides are analysised by infrared spectroscopy.3.UV,HPCE,GC,NMR are applied to analysis the structural elucidation of polysaccharide.4.Antidiabetic activity of Polysaccharides from Alhagi-honey and its mechanism are studied by the experiment of antidiabetic activity in vivo.Results:1.Six homogeneous polysaccharide from Alhagi-honey are obtained with macroporous resin,DEAE sepharose CL-6B ion-exchange chromatography and Sephadex G-100 gel filtration chromatography.Their properties and structures are identified on the basis of methods.The molecular weight of AP1-2 is 1.68×106DA,which is composed of α-L-Arab-(1â†',â†'3)α-L-Arab-(1â†',â†'2)α-L-Rha-(1â†',â†'3)α-D-Man-(1â†'andâ†'6)α-D-Glc UA-(1â†'2);the molecular weight of AP1-3 is 1.74×106 DA,which is composed of â†'3)α-L-Arab-(1â†',â†'5)β-L-Rha-(1â†',â†'5)β-D-Man-(1â†'andâ†'3)α-D-GlcUA-(1â†'2)α-D-GalUA;the molecular weight of AP2-1 is 1.44×106DA,which is composed of α-L-Arab-(1â†',â†'2)β-L-Rha-(1â†',â†'3)α-L-Arab-(1â†',â†'2)β-D-Glc(1â†',â†'3)α-L-Rha-(1â†',â†'5)β-D-Man-(1â†',â†'6)β-D-Man-(1â†',â†'3)β-D-GlcUA-(1â†'β-D-GalUA,â†'2)β-D-Gal-(1â†',â†'3)β-D-Glc-(1â†',â†'6)β-D-Gal-(1â†' and â†'2)β-L-Arab-(1â†';the molecular weight of AP2-2 is 0.98×106DA,which is composed of α-L-Arab-(1â†', â†'5)β-D-Gal-(1â†', â†'5)α-L-Rha-(1â†',â†'2)β-D-Man-(1â†',â†'3)α-D-GlcUA-(1â†'2)α-L-Rha and β-Glc-(1â†'2. The experiment of antidiabetic activity in vivo show that dosing positive drug group and polysaccharides high dose group compared with model group blood glucose have significant differences(P<0.01).Conclusion:1.Six homogeneous polysaccharide are separated and purified,structural analysis and the result shows that the polysaccharides,excepted for AP1-1,are acidic heteropolysaccharide with branches.2.The polysaccharide has potential antidiabetic activity.Its mechanish may not be related to oxidative stress capability.For the first time,the pharmaceutical value of polysaccharide is enriched by amount of researches,the structural elucidation and potential antidiabetic activity of polysaccharide is clarified,which provides a scientific basis for the future rational drug polysaccharide from Alhagi-honey...
