Menopausal syndrome is owing to the decay of the ovaries function, the estrogen level obviously decends, which causes the function disorders of blood vessel, nerve and endocrine system, and which induces the functional uterine bleeding, hectic fever, diaphoresis, the atrophy of cunnus and vagina, osteoporosis and neurovegetative syndromes. With the aging of population struction in China, the age of onset shifes to an earlier time and the disease incidence is ascendant, which severely influence the healthy and quality of life during perimenopausal period and postmenopause occupying one third lifetime.ObjectiveTo investigate the effect of kunbao keli on nervouse activities , the estrogen level, the organ weight, the form of sex organs, and the concent of NE , 5-HT, DA in brain by two kinds of animal model of climacteric syndrome and to study the therapeutic action and the mechanism of kunbao keli on menopausal syndrome.Methods and ResultsPart one The pharmacological study of kunbao keli on fifteen-mouth femal rats1.The effect of nervouse activities on fifteen-mouth femal ratsFifteen-mouth femal rats were chosen as the model animals of menopausal syndrome and were given kunbao keli connectively for twenty-three days in three dosage which were 1.62g/kg, 0.81 g/kg, 0.41 g/kg . and then screen the amounts which the rats moved and made up in box in three minutes. Compared with the climacteric group, the amounts of making up in the treatment group with kunbao keli decreased significantly.2.The effect of memory on fifteen-mouth femal ratsFifteen-mouth femal rats were chosen as the model animals of menopausal syndrome and were given kunbao keli connectively for twenty-six days in three dosage which were 1.62g/kg, 0.81 g/kg, 0.41 g/kg .and then observe the latency entering the dark room and the amounts suffered by electronic. Compared with the climacteric group, the latency entering the dark room prolonged significantly.and the amounts suffered by electronic in the treatment group with kunbao keli reduced significantly.3.The effect of the estrogen level, the organ weight on fifteen-mouth femal ratsFifteen-mouth femal rats were chosen as the model animals of menopausal syndrome and were given kunbao keli connectively for twenty-eight days in three dosage which were 1.62g/kg, 0.81 g/kg, 0.41 g/kg .Levels of serum estradiol(E2) and progesterone(P) were determined by RIA. Weigh the ovary, uterus, renicapsule, and calculate the organ coefficients. Compared with the climacteric group, the levels of serum estradiol(E2), progesterone(P) and the organ coefficient increased significantly .4.The effect of hypothalamic monoaminic transmitters on fifteen-mouth femal ratsFifteen-mouth femal rats were chosen as the model animals of menopausal syndrome and were given kunbao keli connectively for twenty-eight days ,hypothalamic monoaminic transmitters(NE,DA,5-HT) concents were determined by high performance liquid chromatography and electrochemical detector. Compared with the climacteric group, kunbao keli could increase NE content significantly . but kunbao keli couldn't influence the concents of 5-HT, DA significantly.5.The effect of ovary, uterus on fifteen-mouth femal rats by means of HE dyeingFifteen-mouth femal rats were chosen as the model animals of menopausal syndrome and were given kunbao keli connectively for twenty-eight days, the results in HE dyeing showed that, compared with the climacteric group, kunbao keli could promote the growth of ovary and uterus on fifteen-mouth femal rats.Part two The pharmacological study of kunbao keli on ovariectomied mice1.The effect of hypnotize dosage with barbital sodium on ovariectomied miceOvariectomied mice were chosen as the model animals of menopausal syndrome and were given kunbao keli connectively for eleven days in three dosage which were 2.32g/kg, 1.16 g/kg, 0.58 g/kg. compared with the climacteric group, kunbao keli could increase the amonuts of sleep-producing animal, the sleep-pruducing percent is over 50%.2.The effect of hypnotize time with barbital sodium on ovariectomied mice Ovariectomied mice were chosen as the model animals of menopausal syndrome and were given kunbao keli connectively for eleven days. At the eleventh day , i.p barbital sodium 35mg/kg .The results showed that kunbao keli could shorten the sleeping latency and extend the sleeping-time significantly .3.The effect of the estrogen level on ovariectomied miceOvariectomied mice were chosen as the model animals of menopausal syndrome and were given kunbao keli connectively for fifteen days. Levels of serum estradiol(E2) and progesterone(P) were determined by RIA. Levels of serum estradiol(E2) and progesterone(P) have no significant influence in the treatment group with kunbao keli and the climacteric group.Part three The study of antianxiety functionThe health ICR mice were chosen and were given kunbao keli connectively for eight days in three dosage which were 2.32g/kg, 1.16 g/kg, 0.58 g/kg. The results showed that kunbao keli could resist nerve central excitation or convulsion induced by hydromorphinol 20mg/kg. Compared with the model group, kunbao keli could extent the reactive latency of hoist-tail caused by hydromorphinol.Part four The study of memorial functionThe health ICR mice were chosen and were given kunbao keli connectively for ten days in three dosage which were 2.32g/kg, 1.16 g/kg, 0.58 g/kg. The results showed that kunbao keli could resist memory obtain barrier induced by scopolamine. Compared with the model group, kunbao keli could shorten the landing-time , extent the latency entering the dark room and decrease the number of times.ConclusionKunbao keli had certain composed function and has the function of improving memory,increased the estrogen level significantly, improved the function disorders of nerve and endocrine system caused by the estrogen level obviously decend , promoted the growth of ovary and uterus on the model animals of menopausal syndrome;The experimental results showed that kunbao keli could increase the estrogen level ,improved the function disorders of nerve and endocrine system.Compared with the climacteric group, kunbao keli can increase NE content significantly but it cannot influence the concents of 5-HT, DA significantly. From above results ,we could draw a inference that the treatmental mechanism of kunbao keli on menopausal syndrome may be improve the syndrome of neurovegetative disorder induced by the lacing of estrogen,reducd the activity times induced by anxiety,improve the recall function and delay the decrepit of ovary .All of above factors together regulate the body function, enhance the quality of life during perimenopausal period and postmenopause .
