Objective: observing the effect of Dachaihutang on AS rabbits in the development and expression of MCP-1,ICAM-1 mRNA and Protein, to explore the mechanism of Dachaihutang confront with AS. Methods: 40 healthy male New Zealand rabbits, (the weight about 1.5 kg to 2.0 kg) All of them were divided randomly into five groups: normal group,HC model group,Dachaihutang prevention group,Xuezhikang contrast group,Dachaihutang treatment group. After feeding AS rabbits one week, except for Normal group, the other groups were all fed with high-fat diet about ten weeks. At the same time prevention group and Xuezhikang group were also fed with Dachaihutang and Xuezhikang to prevent. After feeding six weeks Dachaihutang treatmentgroup were given with Dachaihutang to treat. Before and after feeding, feeding six weeks and by the end of experiment. measured serum (TC,TG,LDL-C,,HDL-C,CRP). Animals were killed by the end of experiment the sample of arteries were taken measuring MCP-1 and ICAM-1 in immune. Light microscopy were adapted to observe. Results: After threatment the level of TC,TG,LDL-C,CRP in HC model group was markedly higher than in normal group (p<0.01); compared with HC model group, the level of TC,TG,LDL-C,CRP was lower than in prevention group and treatment group (p<0.01); the level of TC,TG,LDL-C,CRP in Xuezhikang group was lower than in HC model group (p<0.01). And obviously differences appeared between prevention group and Xuezhikang group (p<0.01). The expression of Protein in MCP-1 and ICAM-1, there were markly differences appeared among prevention group,Xuezhikang group,HC model group (p<0.01). And obviously differences appeared between prevention group and Xuezhikang group(p<0.01) markly differences appeared between HC modelgroup and treatment group (p<0.05). Condusions: 1. Under the induction of high-fat diet, MCP-1 and ICAM-1 appeared excessive expression, and they can speed artery block. 2. Dachaihutang can reduce the level of serum TC,TG,LDL-C,CRP. 3. Dachaihutang can make the expression of block in MCP-1 and ICAM-1 decrease, means Dachaihutang can restrain the expression of MCP-1 and ICAM-1, it also restrain the development in AS.
