The modernization of traditional Chinese medicine (TCM) is a difficult and formidable project.It is a new tidal wave in the era with informationization,highly developed science and technology.Since China's entery into WTO, people want to go back to the nature.During the process of seeking the comprehensive appraisal of the quality of traditional Chinese medicine (TCM), the fingerprint technique would be the practical and feasible mode to maintain the stability of the production quality and could distinguish the authenticity of TCM. However, with the rapid development of modem hyphenated instruments and related chemometric methods, the analysis of TCM system and quality control though components in TCM become feasible.This research aims at the analysis of Radix isatidis in real TCM system in order to quality control by chemometric methods, and analysis of Radix isatidis and Qinggan Injection by comparing the retention time and UV spectra.In the first chapter, the development of TCM quality control in resent year and its new technology, the application of chromatographic fingerprints, the progress of modem analytical techniques, chemometric methods and the information summarize of Radix isatidis are summarized.Chapter two and three focus on the study of the HPLC and GC chromtographic fingerprints of Radix isatidis.Final determining methods are obtained after the research on preparation method of the sample, solvent and HPLC conditions.The methodological evaluation showed that this method had a good repeatability, precision and stability.The standardization of Radix isatidis data, examination of similarity degree and analysis of chemical pattern were achieved in chapter two and three. What's more, HELP resolution was used to analyses overlapping peaks in order to get pure spectrum and chromatography of each peak, which made the result more accurate and reliable.In chapter four,analysed Radix isatidis and Qinggan Injection sample by comparing the retention time and UV spectra.Relative adscription analysis of components between complex and single medicines of traditional Chinese medicines.A compound medicine of Qinggan Injection sample and its involved single medicines of Radix isatidis were compared for their relevant constituents after the combination of the techniques of hyphenated chromatography and (multicomponent) spectral correlative chromatography, based on the two-dimensional spectrochromatographic fingerprints of the medicinal objects obtained from HPLC-DAD.So it can provide a rapid method for the reach for he complex medicines' quality control.
