Radix Scutellariae is the dried root of Scutellaria baicalensis Georgi.In this thesis, fingerprints quality control methods of Radix Scutellariae were established by HPLC and HPCE, and its super-information characteristic digitized evaluations were presented.The differences between the qualitative similarity and the quantitative similarity were explored,meanwhile,the CZE method was also developed for the quantitative determination of baicalin in Radix Scutellariae. In order to establish a rapid,accurate and scientific method for purification of Radix Scutellariae from the molecular level,provide a theoretical basis for the cultivation and breeding of Radix Scutellariae,DNA fingerprints of Radix Scutellariae was also established by RAPD technical.This study provided new methods for overall quality control of Radix Scutellariae and offered basis for realizing the quantitative function and comprehensive evaluation of fingerprints.1.The index of chromatographic fingerprints(F)was used as quantitative evaluation parameter to choose detection conditions.In order to reflect the water-soluble ingredients,the HPLC-FPS of Radix Scutellariae was established with 30 common peaks under 280 nm,taking the peak of baicalin as referential peak.The qualitative similarities of crude drugs from different resources were evaluated by "Digitized Evaluation System for Super-information Characteristics of the Traditional Chinese Medicine Fingerprints 3.0" while the unified fingerprint of Radix Scutellariae under 280 nm was established.At the same time,this paper also inspected the impact of peak lost to the dual qualitative similarities and the dual quantitative similarities.2.The CEFP of Radix Scutellariae as a novel quality control method was established and evaluated by "Digitized Evaluation System for Super-information Characteristics of the Traditional Chinese Medicine Fingerprints 3.0" while the influences of different background electrolytes, adjusted and pH on the electropherogram were investigated.The CZE method for the quantitative determination of baicalin in Radix Scutellariae was also developed and the bioactive constituent in different batches was determined.The two methods were simple,quick and cost least.They had a good reproducibility and offered new methods for the quality control of Radix Scutellariae.3.The DNA fingerprints of Radix Scutellariae were established by RAPD technical.The impact of different template concentration,different primers concentration,different dNTPs concentration,different TaqDNA polymerase concentration and different temperatures to PCR reaction system were inspected.The electrophorogram of baicalin from different areas were obtained by agarose gel electrophoresis,analysis their genetic differentiation and genetic diversity, and the DNA fingerprints was established.
