The dried tuber of Ophiopogon japonicus is a common used herb for blinding Yinin traditional Chinesemedicine,itwas firstly contained in the "Shen Nong’s Herbal Classic" and ancient books on Chinese herbal medicine all had records about it. It has taste of sweet, slightly bitter, nature of slightly cold, and unto the heart、lung and stomach meridians, Chinese traditional medicine theory includes that thedrug has the effects to nourish yin and tomoisten the lung and ease the mind. It can be used for fever disability allowance, upset thirst and other illnesses. In traditional Chinese medicine, it mainly be used for diseases related with symptoms about yin deficiency of lung、stomach、heart or thermal injury to yin of lung、stomach、heart. Modern clinic and pharmacologic studies found that O.japonicus has effects on enduring hypoxia, anti-arrhythmia,improvingmycoardial contractive, anti-cancer, declining the blood-sugar and so on. The main chemical compositions of Ophiopogon are steroidal saponins、isoflavones、polysaccharides and amino acids, et al. Ithadbeen reported that steroidal saponins and homoisoflavones werethe chief activeingredients of O.japonicus.For the purpose of gettingand riching monomer steroidal saponinschemical constituents ofthis plant, we selected the fresh tubers of Ophiopogon, and carried out various separation methods and systematic investigations of the steroidal saponins. The aimwas to obtain variety of steroidal saponins, so as to provide data and material basis for further study of the pharmacological action and structure-activityrelationshipsof the active ingredient.In this thesis we determined the best craft to extract this herb through preliminary work of accessing, organizing and summarizing variety literatures of Ophiopogon. Then to separate and purifycompounds by means of the D101macroporous resin chromatography、 column chromatography on silica gel、C18reverse phase column chromatography andpreparative High Performance Liquid. Finally got some isolated solid compounds from Ophiopogon.Through checkingphysico-chemical propertiesof compounds and with analysis methods of the’H-NMR、13C-NMR spectroscopic, to identify their structuresand now identification is in progress.Eight structures of these compounds had been identified:Ruscogenin、3-O-α-L-rhamnopyrano-syl-(1'2)-β-D-glucopyranoside(1)、 Ophiopogonin D(2)、Sitosterol(3)、2-Phenylacetamide (4)、stigmasterol3-O-β-D-glucopyranoside (5)、Palmitic acid (6)-methylophiopogonanone A'methylophiopogonanone B(7);and the structures of other compounds were in identification.
