Objective:Cerebrovascular disease is a common and frequently occurring illness,disease with the feature of high disease incidence, high mortality rate and high mutilation rate. A great deal of clinical researches and empirical studies show that acupuncture has unique curative effect on this disease. To approach its' mechanism of action, we carded out this experiment. The SD rats were objects in this experiment. We made rat focal cerebral ischemia model with middle cerebral artery occlusion using a thread block technique. We gave a mark of the neurologic impairment in rat and detected the contents of calcitonin gene-related peptide(CGRP) and neuropeptide Y (NPY) in rats' cornu ammonis and brain mantle respectively by electro-acupuncture(EA) on shuigou(DU 26) acupoint. In this way, we are trying to explore the mechanism of acupuncture therapy in cerebral ischemia.Methods:40 SD rats were selected. Each rat's weight was from 180g to 240g. They were randomly divided into four groups, each group with 10 rats: normal group, sham operation group, model group and model EA group. Normal group rats were not provided treatment. Model group were made focal cerebral ischemia model with middle cerebral artery occlusion using a thread block technique. Model EA group was treated by EA with continuous wave of 120 vices per minute frequency after the rats woke up. The treatment continued for five days, one vice for 30minutes per day. Then, we marked of the neurologic impairment in the rats and detected the contents of CGRP and NPY in the rats' cornu ammonis and brain mantle respectively.Results:1. focal cerebral ischemia rats with right middle cerebral artery occlusion using a thread block technique had behavior disorder to some degree, with levo- circling objective signs, and their left forefoot flection couldn't expand thoroughly. This showed that the CI models succeeded. After treated by EA, neurologic defect objective signs of rat improved obviously. Compared with model group, there was significant difference (P<0.01).2. Compared with sham operated group and normal group, the contents of CGRP in rats' cornu ammonis in model group decreased greatly while the contents of NPY increased obviously(P<0.01). This implyed that the contents of CGRP and NPY in comu ammonis aera had abnormality variation after CI. Compared with model group, the contents of CGRP in rats' cornu ammonis in model EA group increased greatly while the contents of NPY decreased obviously(P<0.01).3. Compared with sham operated group and normal group, the contents of CGRP in rats' brain mantle in model group decreased greatly while the contents of NPY increased obviously(P<0.01). This implyed that the contents of CGRP and NPY in comu ammonis aera had abnormalityvariation after CI. Compared with model group, the contents of CGRP in rats' brain mantle in model EA group increased greatly while the contents of NPY decreased obviously(P<0.01).Conclusion:1. The neurologic impairment of rats with crerbral ischemia improved obviously by EA on shuigou(DU 26) acupoint, indicating that EA can rapair cerebral ischemia damage.2. The contents of CGRP in rats' comu ammonis and brain mantle decreased obviously after CI while the contents of NPY increased, it indicated that CGRP and NPY participate in the brain injured patho-procedure of cerebral ischemia.3. Compared with model group, the contents of CGRP in rats' brain mantle in model EA group increased greatly while the contents of NPY decreased obviously, and it is closely to the contents of normal group. This indicates that EA can adjust the contents of CGRP and NPY in ischemic brain tissue.
