Ovarian cancer is one of the most common gynecological malignant tumors in women, and its mortality is the first. On the pathogenetic early stage,deficiency of characteristic symptom and physical symptom makes the discovery and diagnosis difficult. About 70% of ovarian cancers enter its advanced stage when they are diagnosed.Compared with other malignant gynecological tumors,ovarian cancer is charactered with high-malignancy, early faraway-transferring and being removed difficulty. Recent studies have revealed the CD44 variant 6 (v6) molecule and PCNA have been noted as a marker for tumor metastasis and prognosis in several tumors.CD44 is an adhesion molecule involved in cell-to-cell and cell-to-matrix interactions. This transmembrane glycoprotein exists in either standard or variant forms, originated by alternative splicing. PCNA (proliferating cell nuclear antigen), originally characterized as a DNA polymerase accessory protein, functions as a DNA sliding clamp for DNA polymerase delta and is an essential com- ponent for eukaryotic chromosomal DNA replication.PCNA has applications as a marker for DNA synthesis. It is of interest that proteins involved in cell cycle regulation may also exhibit PCNA binding activity. It has clinical sense to find influential factors and relatively mechanism of ovarian carci- nomas biological behaviour, and to find method and index used in diag -nosing early, detecting recurrence and evaluating prognosis.Objective:To study the expression of CD44v6 and PCNA in epithelial ovarian carcinoma, and the relationships of expression with the curative effect of chemotherapy and prognosis.Methods: To detect the protein expression of CD44v6 and PCNA gene in 20normal,20 benign and 23 malignant epithelial ovarian tumor tissue with SABC method of immunohistochemistry techniqueResults:1.Positive expression of CD44v6 in malignant(78.26%)was higher than the expression in benign(15%)and normal tissues(0%)(P<0.05).2.There were no significant correlations between the expressional grade of CD44v6 and the clinical stage(P>0.05), No significant correlation was found between the expressional grade of CD44v6 and ascites(P>0.05 ), No significant correlation was found between the expressional grade of CD44v6 and lymph node metastasis(P>0.05 )3.Positive expression of PCNA in malignant(86.96%) was higher than the expression in benign(10%)and normal tissues(0%)(P<0.05).4.There were no significant correlations between the expressional grade of PCNA and the clinical stage (P>0.05 ),No significant correlation was found between the expressional grade of PCNA and ascites (P>0.05 ), No significant correlation was found between the expressional grade of PCNA and lymph node metastasis(P>0.05 )Conclusion : The detection of CD44v6and PCNA may predict the curative effect of chemotherapy and prognosis in epithelial ovarian carci- noma.PCNA may be used as an important predicting marker for the prognosis of patients with ovarian epithelial neoplasmas.The expression of PCNA could be used as a valuable indicator of the biological behavior and prognosis of ovarian epithelial cancer.
