The Rhizoma et Radix Baphicacanthis Cusae collected in 2005 editions the Chinese Pharmacopoeia originate from the acanthaceae plant of Baphicacanthus cusia (Nees) Bremek rhizome and root, its nature is cold, its taste is bitter, and it can improve the function of the stomach, having fairly good effect of clearing heat and detoxication, and the antibacterial sterilization effect. On clinical it is used in seasonal contagious disease, such as the erysipelas, flu, epidemic cerebrospinal meningitis. Rhizoma et Radix Baphicacanthis Cusae is a perennial herb, scatters broadly in southwest of China, the South China and the Eastern China, its ground parts (stem, leaf) can be used as medicine directly or processing the dried indigo vat froth into medicine, it has higher utilization ratio. At present, taking Rhizoma et Radix Baphicacanthis Cusae as responsibility raw material in traditional Chinese medicine preparation, there mainly have: Compound prescription south isatis root piece, Compound prescription south isatis root granular formulation, compound prescription south isatis root inoculation fluid and so on.However, the Rhizoma et Radix Baphicacanthis Cusae medicine source mainly depends wildly at present, factors such as the long vegetative cycle , the low output, and the excessive excavation all lead to the wild resources to reduce suddenly; there are also massively heterogeneous false which come from the same genus but different species in market, all that seriously influence the normal clinical medication need and the traditional Chinese medicine production which takes Rhizoma et Radix Baphicacanthis Cusae as responsibility raw material.For further study on the standardized planting of Rhizoma et Radix Baphicacanthis Cusae, this article is about the observation and the research of Rhizoma et Radix Baphicacanthis Cusae 's biology characteristic, mainly includes: Literature search, status of resources and shape characteristic research, growth habit research, ecological environment condition research, indirubin content determination methods research, and harvest method research. The concrete result is as follows:1. The massive material about the study on Rhizoma et Radix Baphicacanthis Cusae as medicine as well as material related research have been consulted. The summary mainly involve the research and the progress of the Rhizoma et Radix Baphicacanthis Cusae's book on Chinese medicine research, base original appraisal, character and micro appraisal, chemical composition, pharmacology toxicology, cultivation research and so on.2. The resources status of Rhizoma et Radix Baphicacanthis CusaeIn the combination of consulting literature and on-the-spot investigation, we discovered that wild Rhizoma et Radix Baphicacanthis Cusae mainly distribute in South China, Southwest, partial provincial areas of East China as well as Taiwan; But the artificial cultivation of Rhizoma et Radix Baphicacanthis Cusae have not been plantdted massively, only manual planter in small areas of Guizhou, South Chekiang ;Most of the Rhizoma et Radix Baphicacanthis Cusae sold in the market are heterogeneous false which originate from the same genus but different species such as Strobilanthes pentstemonoides, Strobilanthes oliganthus Miq, Pteracanthus guangxiensis, Serobilanthes divaricatus . The medicinal resources are relatively short.3. Study of biology characteristic of Rhizoma et Radix Baphicacanthis Cusae3.1 The morphologic characteristics.Compare the morphologic characteristics of wild and the family planting ones, detect that obvious difference between with them, the main differences are: The family planting one's stem erectness, sturdy, the rhizome is short and eribble, and many fibrous root; Wild one's stem always lodging, exquisite, the rhizome is long and exquisite,and few fibrous root.3.2 Study on growth and development habit of Rhizoma et Radix Baphicacanthis Cusae.Objection: Inspects the height and branching number in different time to comprehend the growth and development habit of Rhizoma et Radix Baphicacanthis Cusae.Method: Selects the experimental field, about 500 elevation meters, in good ventilates and draining water in Shi Zhengzhen Pingyuan County. Breeding spire by vegetative form, then transplant to the open ground, shine the straight light for 6 hours every day. 10 plants are selected stochastically to carry on observation by license tracing. Each month measure the height of plant, the mean value of each total branching number in the 1 year after being planted, draw the curve diagram of dynamic state changes.Result and conclusion: In the observation period, the trend of height and total number of branching is upgrade.The plant raise up quickly after been field planting until August, in September, the speed slow down until the almost stop; the branching number gradually increases after been field planting until September, during the September to December increase obviously quickly, after December, branching number obvious reduce. Obviously its peak of vegetative growth is after been field planting until August, the peak of reproduction growth is during September to December.3.3 Study of condition of biogeocoenosisThe suitable temperature: 15-30℃, the growth peak: spring and fall. It is half shade plant, like sunlight, warm, also can bear overcast, certain time of straight light illumination is advantageous to its growth, basely can bear cold, also like moist, but do mot like dropsy. It has strong compatibility to the soil, should be planted in loose, fertile, good draining water neutrality or alkalescence sandy loam and loam.In order to inspect the effect of illumination to the growth of Rhizoma et Radix Baphicacanthis Cusae, this article study the effect of different illumination condition to the height and the number of branching :Objection : Inspects the influence of different illumination condition to Rhizoma et Radix Baphicacanthis Cusae the height and the number of branching.Method: Refers to the previous experiment to select field and plants, provides shade to the plant by planting shade plant or exploitation relief, establishes for the no illumination group, 4 hours illumination group, 6 hours illumination group, adopts the same methods of previous experiment on the plante and the measure.Result and conclusion: The illumination make great effect to the growth of Rhizoma et Radix Baphicacanthis Cusae, the height and the number of branching displays that: 4 hours illumination group >6 hours illumination group > the no illumination group, obviously that certain time's straight light illumination is advantageous to the growth of Rhizoma et Radix Baphicacanthis Cusae, but strong illumination will suppress it. Rhizoma et Radix Baphicacanthis Cusae can be planted under the suitable illumination condition in the open ground by artificial.4. Determination method study of indirubin in Rhizoma et Radix Baphicacanthis CusaeObjection: Inspects the effect of different resolver and extraction method that determines the indirubin Rhizoma et Radix Baphicacanthis Cusae contents, screening scientific, convenient method.Method: Respectively by the benzine, the chloroform, the absolute ethyl alcohol, by three different extraction method :rope, the backflow, the supersonic, with the C18 , moving phase: methyl alcohol - 0.1% acetic acid solution (80:20), flow: 1.00mL/min wave length: 290nm. Result and conclusion: Indirubin has good relationship in 5.375ng~86ng (r=0.9994), extracted by the absolute ethyl alcohol backflow has the best effect; Indirubin stables in 24 hours, redundant specimen handling RSD=1.28%, parallel experiment obtained RSD=2.33% (n=5), the returns-ratio is 96.22%, RSD=1.65 (n=5). Each target of this method meets the requirement of instrumental analysis.5. Study on the reasonable collection period of Rhizoma et Radix Baphicacanthis CusaeObjection: determine the content of indirubin in different recovery periods to determine the best recovery period.Method: Select the groups: after planting 8 months no illumination group, 4 hours illumination group and 6 hours illumination group. each group choose 10 immediately. surveys each's weight, and determines the content of indirubin by HPLC, draw the diagram of curves of dynamic change.Result and conclusion: In the observation period, output of Rhizoma et Radix Baphicacanthis Cusae rise slowly , the content of indirubin achieves the peak during December and January, the output and the content even present: 4 hours illumination group >6 hours illumination group > the no illumination group. Therefore, Rhizoma et Radix Baphicacanthis Cusae should be plant under 4 hours straight light illumination, the best recovery period is the end of November to the beginning of December.6. Study of best harvest part of Rhizoma et Radix Baphicacanthis Cusae Objection: to find out its best harvest part by measuring of indirubin content indifferent original parts of Rhizoma et Radix Baphicacanthis Cusae.Method: Take the leaf, the stem, the root and the rhizome part of Rhizoma et Radix Baphicacanthis Cusae separately, then smash them, and determine the content of indirubin in each part by HPLC .Result and conclusion: according to the result, the content of indirubin in each part of Rhizoma et Radix Baphicacanthis Cusae is different, to the leaf is the highest, to the stem is next, to the root and the rhizome part are the lowest. For the present market, we suggested to taking the root and the rhizome and the ground mature stem of Rhizoma et Radix Baphicacanthis Cusae as the harvest part by further chemistry and the Pharmacology study.
