ObjectiveAs the basis of Chinese Pharmacopoeia (2010), establish the quality evaluation method of the leaves of Baphicacanthus cusia (Nees) Bremek., and then set up a quality standard of it. What we have done is for the leaves of Baphicacanthus cusia (Nees) Bremek. to be collected by state standards and local standards, and it will help to guide the safety and effectiveness in clinic.MethodsQuality standard of the leaves of Baphicacanthus cusia (Nees) Bremek. was established according to Chinese Pharmacopoeia (2010), concrete research content including morphological identification, microscopic identification, thin layer chromatography (TLC), and water content, total ashes, acid-insoluble ashes and extractives. In addition, establish a research method of HPLC fingerprint for the dichloromethane extractive from the leaves, and develop a method for determination content of five index components base on the fingerprint.Results1. Pharmacognosy study on the leaves of Baphicacanthus cusia (Nees) Bremek. The authentic and adulterants can be identified by methods of morphological identification, microscopic identification and TLC. While the cultivated and wild samples also can be identified by methods except TLC.2. Determination of the leaves of Baphicacanthus cusia (Nees) Bremek.’s conventional inspection Conventional inspection of the leaves of Baphicacanthus cusia (Nees) Bremek. was established according to Chinese Pharmacopoeia (2010), including water content, total ashes, acid-insoluble ashes and extractives, in order to establish the maximum limit. 3. Study on HPLC fingerprint of the dichloromethane extractive from the leaves of Baphicacanthus cusia (Nees) Bremek. The fingerprint of the dichloromethane extractive from the leaves of Baphicacanthus cusia (Nees) Bremek. was established by HPLC, and the method was stability, precision and reproducibility. Twenty batches of leaves’fingerprint similarity was greater than0.9by similarity calculation, in addition, the samples can not be divided according to the origin or clustering by cluster analysis, illustrate the quality of the leaves from different regions was similar, and the wild and the cultivated also no significant difference.4. Determination of five index components’content in the leaves of Baphicacanthus cusia (Nees) Bremek. On the basis of HPLC fingerprint of the dichloromethane extractive from the leaves of Baphicacanthus cusia (Nees) Bremek., the simultaneous determination of five index components in thirty samples was established, including benzoyleneurea,4-hydroxyquinazoline,2-methyl-4(3H)-quinazolinone, tryptanthrin and indirubin. on the southern Folium collected for testing of medicines, the method is stable and reliable.5. Completed the drafting of the quality standard of the leaves of Baphicacanthus cusia (Nees) Bremek.ConclusionEstablish a quality standard of the leaves of Baphicacanthus cusia (Nees) Bremek., for quality control of it with modern technology and methods of qualitative and quantitative analysis and evaluation, also for the leaves of Baphicacanthus cusia (Nees) Bremek. to be collected by state standards and local standards, and to guide the safety and effectiveness in clinic.
