In this study,the medicinal components in leaves,stem and root of Baphicacanthus cusia(Nees)Bremek from 14 different germ plasm resource in Fujian province were detected by HPLC-UV,and the HPLC fingerprint in different parts of Baphicacanthus cusia(Nees)Bremek were studyed.To understand the effect of the environment and planting age on the results of quantitative analyses,the variation of medicinal ingredients in Baphicacanthus cusia(Nees)Bremek from fourteen different regions and in Baphicacanthus cusia(Nees)Bremek from three regions in different growth season and in different planting age was analysised.The results are as follows:1 The HPLC fingerprint method for the determination the medicinal components in Baphicacanthus cusia(Nees)Bremek in Fujian was established.Chromatographic conditions were:WatersC18 column(5?m,4.6÷250mm),the mobile phase was acetonitrile-0.05%phosphoric acid(gradient elution),flow rate was 0.8mL/min,the detection wavelength was 254nm,the injection volume was 20?L,column temperature was 25 ? and the analysis time was 85min.2 The HPLC fingerprint in leaf,stem and root of Baphicacanthus cusia(Nees)Bremek were obtained.Sixteen common peaks(the 9th peak was the tryptanthrin,the 11th peak was the indigo and the 12th was indirubin)in the fingerprints of Baphicacanthus cusia(Nees)Bremek leaf were determined.The 13 common peaks(the 8th peak was tryptanthrin,the 10th peak was indigo and the 11th was indirubin)in the fingerprints of Baphicacanthus cusia(Nees)Bremek stem were determined.The 11 common peaks(the 7th peak was indigo and the 8th was indirubin)in the fingerprints of Baphicacanthus cusia(Nees)Bremek root were determined.Fingerprint analysis showed the similarity between the HPLC fingerprint in the same part(leave,stem,root)from different Baphicacanthus cusia(Nees)Bremek and the difference between the different parts from the same sample.3 There were significantly differences between the contents of medicinal ingredients in Baphicacanthus cusia(Nees)Bremek from different germ plasm resource in Fujian province.The contents of indirubin in aboveground and underground that from BY,YC,BZ,XZ were higher than other germ plasm resource.The contents of indigo that from PT,BZ,YQ,XZ were higher than other germ plasm resource;The contents of polysaccharides that from YC,YQ,XZ were higher than other germ plasm resource.4 There were significantly differences between the contents of medicinal ingredients in Baphicacanthus cusia(Nees)Bremek in different seasons.The contents of indirubin and indigo increased first and decreased afterwards,and reached the highest in autumn.But the contents of polysaccharides were up to the highest in summer.5 There were significantly differences between the contents of medicinal ingredients in Baphicacanthus cusia(Nees)Bremek in different planting age.the contents of indirubin and indigo in Baphicacanthus cusia(Nees)Bremek were relatively high in the first year.And the contents of polysaccharides were relatively high at the second or third year.
