In order to probe the chemical constituents of the root of Isatis indigotica fort (Radix isatis) and Baphicacanthus cusia(nees) bremek (Rhizoma et radix Baphicacanthus) and compare the officinal portion of Isatis indigotica fort with that of Baphicacanthus cusia(nees) bremek, we had systemically extracted ,isolated and indentified the root of Isatis indigotica fort and the root of Baphicacanthus cusia(nees) bremek, and had also researched the fingerprint spectrums of them by HPLC. Furthermore, we had studied the character and microstructure feather of them .The constituents isolated and identified from Isatis indigotica fort include sterols, sterolines, indoleorganic acids and disaccharides. They were 1-methoxy-3-acetic acid, Indole, daucosterol, adenosine,plamitic acid,β-Sitosterol, 16-methyl-heptadecanoic acid,Indigotin,Indirubin,Sucrose and Pentadecanoic acid. 1-methoxy-3-acetic acid was obtained from the root of Isatis indigotica fort for the first time.The constituents isolated and identified from Baphicacanthus cusia(nees) bremek were sterols, pentacyclictriterpenoids, indole alkaloids,phenylethane glycosides and disaccharides. They were Stigmasterol-3-O-glucopyranoside,Stigmasterol-3-O- glucopyranoside,Lupeol, [(2-(3,4-dihydroxyphenylethyl ) ) -3O-α-L-rhamnopyranosyl-(1→4)-(4-O-caffeoyl)-β-D-glucopyranosi de] ,Indigotin,Indirubin,Sucrose,adenosine andβ-Sitosterol) . Stigmasterol-3-O- glucopyranoside was obtained from the root of Isatis indigotica fort for the first time.At the same time,proper method for the determination of HPLC fingerprint spectrum of the root of Isatis indigotica fort and the leaf of Isatis indigotica fort was established and up to the technical demands for TCM fingerprint spectrums. HPLC fingerprint spectrums of different places and source of the root of Isatis indigotica fort and the leaf of Isatis indigotica fort was determined and 12 common fingerprint peals was ascertained. Similarities of the fingerprint spectrums were compared and showed the quality difference for the different plants. The fingerprint spectrums of the root of Isatis indigotica fort and the leaf of Isatis indigotica fort were compared each other and it showed that their components were different. The result was different with that of the studies on the chemical constituents. The similar is lower than 0.6.Through the studies on the constituents, HPLC fingerprint spectrums, medicinal purposes spot character and microstructure research of Isatis indigotica fort and Baphicacanthus cusia Bremek, we find that the two kinds of medical plants were completely different. They come from two branches of medical plants and the blood relationship is farther.The chemical constituents, HPLC fingerprint spectrum, microstructure research of medical character of Isatis indigotica fort and Baphicacanthus cusia Bremek were studied in the paper, and it was showed that the medical plants are completely different, although their medical effectiveness and pharmacological action were similar, their chemical composition was completely different. The two medicines should be used distinguishedly to treat many diseases in the clinic.
