From The Monoamine Neurotransmitters And Their Receptors To Explore Yinao Jieyu Side Of The Intervention Mechanism Of Depression After Stroke

AIM: According to the etiopathogenisis theory of "endogenous mechanism" and "reactivity mechanism" and Chinese medicine theoty of lossing support of mind caused by renal dificency,depressed vital energy and stasis of phlegm and stagnant blood about poststroke depression(PSD) to duplicate the rat model with PSD,and investigate the pathogenesy of PSD and the effects of Yinao Jieyu prescription on PSD from the points of monoamie transmitters and their receptors according to the pathogenesy hypothesis of monoamine transmitters and receptor of depression.METHODS: Male Wistar rats were randomly divided into normal group,stroke group,model group,treatment group and control group according to the scores of Open-Field test.Rats in the normal group received no intervention, those in other four groups were duplicated into focal ischemia first,and they were made into PSD model except those in the stroke group,and then they received intragastric administration of double distilled water,fluoxetin and Yinao Jieyu prescription respectively once a day for 6 weeks from the 1st day after the establishment of model.After that the behavioral evaluation were done with consumption of sucrose water,Open-Field test and Morris water maze ,and then the brain samples were taked with different ways for mesurement of the index (1) the contents of norepinephrine(NE),dopoamne(DA),5-hydroxytryptamne(5-HT) and their metabolites in the left anterior cerebral cortex and hippocampus were determined with the high performance liquid phase-electrochemical method;(2)the expressions of α2A adrenaline receptor (a 2aAR), D2 Dopamine receptor (D2DR), 5-hydroxytryptamine 1A receptor (5-HT1aR) and tyrosine hydroxylase (TH) of the paraffin sections or frozen sections were done with SABC method immunohistochemistry,and optical density and area density were measured with image analysis at the left anterior cerebral cortex and hippocampus.RESULTS: (1)Behavioral evaluation: Compared with those in the normal group and stroke group,the consumption of sucrose water ,the scores of upright and horizonal activities of the model group were decreased significantly,the, the results of Morris water maze showed that the mean escape latency of the model group were prolonged,initial angle were larger,the times of crossing the platform, percentage time and distance in the target quadrant to the tatal swimming time and distance were reduced significantly in the model group (P<0.05;P<0.01);compared with those in the model group,the consumption of sucrose water,the scores of horizontal and upright activity of Open-Field test and the mean escape lantencies of the Morris water maze on the fourth day reduced,the initial angle was smaller,the times of crossing the platform reduced and the percentage of time and distance in the target quadrant to the total time and distance were increased significantly in the treatmentgroup and control group (P<0.05;P<0.01).(2)Measurement of contents of monoamine transmitters and expression of TH: In the anterior cerebral cotex,the 5-HT,5-HIAA contents in the model group were lower than those in the normal group and stroke group(P<0.01),and the content of NE was lower than that in the normal group(P<0.05);compared with those in the model group.the contents of 5-HT and 5-HIAA in the control group (P<0.05),5-HT and NE in the treatment group were higher(P<0.05;P<0.01).In the hippocampus,the contents of 5-HT,5-HIAA and NE in the model group were lower than those in the the normal group and stroke group (P<0.05;P<0.01),and DA was lower than that in the normal group;compared with those in the model group,the contents of 5-HT and 5-HIAA in the control group(P<0.05;P<0.01),DA and NE in the treatment group increased significantly(P<0.05;P<0.01).The optical density and area density of the expression of TH at hypothalamus in the model group and stroke group were significantly lower than those in the nornal group((P<0.05;P<0.01)),and they were higher in the control group and the treatment group than those in the model group(P<0.05;P<0.01).
