Insomnia is one of the general diseases in sleep disorders with serious influence on patients'working and living. The medicines currently used for Insomnia in Clinical like Benzodiazepine, shows good effect on promoting patients to sleep, but makes the patients feel dizzy, burnout, sleepy and unclear-headed after sleep, especially in the initial stage of awaking, and long-term use will cause the Withdrawal Syndrome and Drug Dependence, so the developing the safe, effective drugs with less Side effects and Adverse effects is imperative. Traditional Chinese Medicine (TCM) has been used to treat insomnia for thouands years, which also shows a satisfied effect but lack of efficiency; and the great progress in the modernization of TCM makes it possible to develop some new drugs for treatment of insomnia based on the some effective components from TCM.Panax ginseng C.A.Meyer is widely used in Traditional Chinese Medicine for the treatment of insomnia. Ginsenoside Rb1 (GsRb1) is a main effective component of Ginsenoside, extracted from the stem, root, leave, bud of Panax ginseng C.A.Meyer. The previous studies suggested that the GsRb1 had an inhibitory effect on the central nervous system (CNS).In the study, we are trying to evaluate the effect of GsRb1 on sleep through Sleep ECoG (Electrocorticogram) and Morris Water Maze Test. From the ECoG of Rat, it showed that GsRb1 significantly enhanced the Slow Wave Sleep (SWS), especially the dose (20mg/kg) significantly increased the Single Total Sleep Time, and the dose (10mg/kg) especially showed the enhaced effect on SWS; the analysis of Power Spectral showed the similar results of GsRb1 on SWS.Secondly, we designed a new method to test the side effect of Medicine on the initial stage of awaking, and then we could evaluate the effect of Medicine and the quality of sleep from 2 processes, sleep and awaking. The results showed the 3 doses of GsRb1 significantly decrease the Escape Latency, and also increased the performance of Memory function, which suggested the GsRb1 may not have a side effect like sleepiness and dizziness.In general, the result showed GsRb1 is not dose-depending, and the dose (20mg/kg, 10mg/kg) have the good potential to improve the quality of sleep.
