Objective:To investigate the expression of tumor supresssr gene PT EN and apoptosis inhibitor gene Survivin in human carcinoma of endometrium and study its clinical-pathological implication and its clinical significance.Methods:The expression of PTEN,NF-κBp65 and Survivin were detected by immunohistochemistry SP method in 68 cases of carcinoma of endometrium 21 of atypical hyperplasia of mucosa,33 of adjacent normal mucosa and 46 cases the metastasis lymph nodes.Results:1(1)Expression of PTEN in the tumor cells is below than in the Atypical hyperplasia,and the proliferative phase of endometrium.There was a significant positive correlation between carcinoma of endometrium,Atypical hyperplasia and proliferative phase of endometrium(P<0.05).(2) the PTEN protein expression cut down step by step with the increase of clinical stage and the decrease of the differentiation2(1) Expression of Survivin in the tumor cells is higher than in the Atypical hyperplasia,and the proliferative phase of endometrium.There was a significant positive correlation between carcinoma of endometrium,Atypical hyperplasia and proliferative phase of endometrium (P<0.05).(2) the Survivin protein expression to rise gradually with the increase of clinical stage and the decrease of the differentiation.3(1) Expression of NF-κBP65 in the tumor cells is higher than in the Atypical hyperplasia, and the proliferative phase of endometrium.There was a significant positive correlation between carcinoma of endometrium,Atypical hyperplasia and proliferative phase of endometrium(P<0.05).(2) the NF-κBP65protein expression to rise gradually with the increase of clinical stage and the decrease of the differentiation.Conclusion:1.Reducing expression of PTEN protein occurs commonly in carcinoma of endometrium and correlates with tumor stage,histological poor differentiation and lymph node metastasis.PTEN gene play an important role in the occurrence and development of carcinoma of endometrium.It is suggested that P'fEN can be a useful marker for predicting invasion and met stasis ability of carcinoma of endometrium.2 The abnormal expression of NF-κBP65 protein occurs commonly in carcinoma of endometrium and correlates with tumor stage,histological poor differentiation and lymph node metastasis.NF-κBP65 gene play an important role in the occurrence and development of carcinoma of endometrium.It is suggested that NF-κBP65can be a useful marker for predicting invasion and met:::stasis ability of carcinoma of endometrium.3.The abnormal expression of Survivin correlates with the occurrence and development of carcinoma of endometrium.Survivin was over-expression in carcinoma of endometrium.Expression of Survivin had a significant relation with the invasion depth,lymph node metastasis,TNM stage.Survivin is an important prognostic indicator of carcinoma of endometrium4.There are negative correlation between PTEN and Survivin,NF-κBp65 through detecting they in carcinoma of endometrium.They take part in the occurrence and development of carcinoma of carcinoma of endometrium.Combined detection can do early diagnosis and estimate prognosis.
