Gehua Prevention Of Alcoholic Liver Injury In Experimental Studies

Alcoholic liver disease is a common clinical disease,caused by long-term alcohol abuse.ALD is a main reason of liver-related disease and death in western countries.In china,by the development of living level,the number of drinker increased greatly and the incidence of ALD increased year by year.Although modern medicine are recognizing and researching ALD deeply,the therapeutic tool is limited,it is short of drugs which is safe and effective.The effects of Chinese materia medice is mildly and have little side effects,it have some superiority in prevention and treatment of this disease.This research aims at applying the theory of TCM combined with the modern scientific method to uncover the theory and experience about ALP,discussing the function mechanism of traditional medicine Flos Puerariae preventing ALD,enlarging the clinical application of Flos Puerariae,providing reference for the clinical usage of Chinese materia medice reasonably,safely and efficiently.1.Study of literatureWe collected considerable literature of ALP from ancient period to present period,and collected clinical reports published in medical magazines in 1987-2007 which treat ALD.Then discussed the special feature about the name, etiopathogenisis,diagnosis,treatment and prescription by differentiation of syndromes of TCM about ALD.The result of analysis is that the pathogenesis of ALD results from the mutual contention of qi,blood,phlegm and dampness.Fortifing the spleen and soothing liver,regulating qi and activating blood and resolving phlegm and dispelling dampness is the main treating method.We should reinforce the healthy qi in the advanced stage due to exuberance of pathogen with decline of the healthy qi.Blood-activating and stasis-resolving medicina,dampness-draining diuretic medicinal,dampness-resolving and qi-moving medicinal are common treating medicine.Flos Puerariae is a traditional reliving alcohol medicine,modern pharmacoresearch indicates Flos Puerariae may protect the liver,this research aims at observing the effect of Flos Puerariae on ALD and study its function mechanism.2.Empirical studyOn the basic of searching the newest advancement about ALD,empirical study about Chinese material medicine preventing and curing ALD,we design the following experiments:2.1 Experiment study of weight and liver index:The ALD model rats was duplicated by feeding alcohol,we observe the weight and liver index,the results demonstrate extraction of Flos Puerariae can improve the drunk condition,keep the weight increase and decrease the liver index.2.2 Effects on the activity of serum alanine aminotransferase(ALT),aspartate aminotransferase(AST):we observe the level of ALT and AST in the blood serum,the results demonstrate extraction of Flos Puerariae can reduce the level of ALT and AST in the blood serum,and stabilize the cellular membrane.2.3 Effects on the activity of serum alkaline phosphatase(ALP),the content of total protein(TP)and albumin(ALB):we observe the level of ALP,the content of TP and ALB in the blood serum,the results demonstrate extraction of Flos Puerariae may reduce the level of ALP,increase the content of TP and ALB.2.4 Effects on lipid peroxidation:we observe the activity of liver superoxide dismutase(SOD),the content of liver maleic dialdehyde(MDA)and glutathione(GSH) in the liver tissue,the results discover that extraction of Flos Puerariae can stop the exhaustion of SOD and GSH,drop the content of MDA,the results suggest that it can inhibit lipid peroxidion.2.5 Effects on alcohol dehydrogenase(ADH):we observe the activity of ADH in the liver tissue,the results demonstrate extraction of Flos Puerariae can activate the ADH.2.6 Effect on the alteration of liver pathological shape:we observe the alteration of liver pathological shape,the results suggest that extraction of Flos Puerariae can improve the adipose degeneration and inflammation of liver tissue,and alleviate liver' s pathological injury.Based on these above results,we speculate that Flos Puerariae can stabilize the cellular membrane,improve the liver function,inhibit lipid peroxidion, activate the ADH,alleviate liver' s injury.
