Persimmon Calyx is the dry calyx of Diospyros kaki L.,which is widely distributed in China.It is one of the widely used traditional Chinese medicines,which has been used for years in the treatment of stroke or syndrome of apoplexy in clinics in China to improve the outcome of ischemia stroke.Several active components have been reported on Persimmon Calyx.However,less systemic information between clinic effect and active component can be found in the literature in our country.The aim of this study,therefore,one is to supply substantial bases for further study relationship between bioaetivity and structure,another is to increase the economic of the enterprise and make full use of the rsource.Focusing on chemical structure and the method of extracting and purification of active components,we carried out following studies.Dry plant powdered was extracted with EtOH at room temperature.Removal of solvent by evaporation,the combined extracts(1000g)were suspended in H₂O,After filtrate,The aqueous layer was partition edagain with CHCl₃ and n-BuOH.The CHCl₃ extract was ehromatographed on sitica-gelcolumn,using mixtures of CHCl₃ and MeOH as eluent to give components 1-2.Another fraction was chromatographed on sephadex LH-20 with MeOH -H₂O(20-80%)as solvent to give components 3-5.Finally,five compounds were separated and were identified as follows,Ⅰ: Oleanolie acid((3β)-3 -hydroxy-olean -12 -en -28 -oic acid);Ⅱ:Ursolic acid (3β-hydroxy-urs-12-en-28-oic acid);Ⅲ:Gallic acid;Ⅳ:Ethyl gallate;Ⅴ:Quercetin. Among them,Ethyl gallate was isolated from Persimmon Calyx for the first time.We found that the oleanolie acid and ursolic acid are main bio-active components in Persimmon Calyx.Oleanolie acid and Ursolic acid are pentacyclic triterpenoid compounds,which are geometrical isomers of each other.The research expressed that the design of process route is reasonable,the method is proper and the purity and recovery yield of product come to higher level.
