Objective:To investigate the expression of MMP-26 and integrinανβ3 mRNA in the cultured eutopic endometrium of adenomyosis and in normal endometrium of uterine leiomyoma.To make out the function of mifepristone on the expression of MMP-26 and integrinανβ3 mRNA. Furthermore,to elucidate the mechanism of adenomyosis.Methods:1.Culture the cell of eutopic endometrium in adenomyosis and uterine leiomyoma.2.Observe the expression of MMP-26 and integrinανβ3 in eutopic endometrium of adenomyosis and in normal endometrium.We detect the mRNA expression of MMP-26 and integrinανβ3 through RT-PCR.3.Investigate the function of mifepristone.Find the effective concentration of mifepristone by MTT.Then detect the mRNA expression of MMP-26 and integrinανβ3 after the treatment of mifepristone.Results:1.The expression of MMP-26 in eutopic endometrium of adenomyosis is significantly higher than in normal endometrium.2.The expression of integrinανβ3 in eutopic endometrium of adenomyosis is significantly lower than in normal endometrium.3.There were significant differences in the groups of 5mg/L and 2.5mg/L.Mifepristone inhibited the growth of endometrium of adenomyosis.Mifepristone induced the expression of MMP-26 downregulated and the expression of integrinανupregulated in eutopic endometrium of adenomyosis.But the expression of integrinβ3 has no apparent difference between the group treated with mifepristone and the group without the treatment.Conclusion:1.The upregulation of the expression of MMP-26 in eutopic endometrium of adenomyosis is correlated with increased invasiveness and the progress of adenomyosis.2.The decreased expression of integrinανβ3 in eutopic endometrium of adenomyosis might be correlated with the lower adhesiveness of adenomyosis.3.Mifepristone inhibits the growth of endometrium of adenomyosis.This suggests mifepristone might have some functions in the therapy of adenomyosis.
