| Backgrounds and Objectives:Transforming growth factor-beta (TGF-β) is a pleiotropic growth factor that plays a critical role in the development of embryo, modulating cell growth and differentiation ,the balance of extracellular matrix and tumorigenesis and invasiveness. Generally believed, there are three isotypes of TGF-βin mammalian animals : TGF-β1,TGF-β2,TGF-β3。Relatively , there are three kinds of its receptor : TÎ’R-I,TÎ’R-â…¡,TÎ’R-â…¢. Evidence indicates that TGF-β1 plays roles in transfering signals by combining with transforming growth factor-beta receptorâ…¡(TÎ’R-â…¡). Research discovers in vivo and vitro, expression changes between TGF-β1 and TÎ’R-â…¡in diverse malignant tumour .Accordingly, TGF-β1and TÎ’R-â…¡are involved in malignant tumour .Oral squamous cell carcinoma is one of the most frequent malignant tumour in which tongue cancer is the most frequent. The incidence is working up recently. Pristine tongue cancer can provoke lymphatic metastasis because tongue has redundant blood and lymphokinesis. Accordingly, immunohistochemical streptavidin/peroxidase technique will be used to detect the expression of TGF-β1and TÎ’R-â…¡in this experiment to reveal further the relationships between TGF-βsignal pathway and the features of clinical pathology of squamous cell carcinoma of tongue and to provide theory evidence for clinical therapia.Methods:Immunohistochemical streptavidin/peroxidase technique was used to detect the expression of TGF-β1and TÎ’R-â…¡in archival tongue samples including 40 cases of squamous cell carcinoma and 10 cases of normal tongue tissue. Furthermore,Statistics analysis for SPSS13.0 was used to investigate the relationships between TGF-βsignal pathway and the features of clinical pathology of squamous cell carcinoma of tongue.Results:1. The expression of TGF-β1 :â‘´There were 23 positive cases in 40 samples of squamous cell carcinoma of tongue ,and the positive rate was 57.50%. In comparison with normal tongue tissue the difference was significant(P<0.05)。⑵The positive expression of TGF-β1 among the different age groups or different histological grades showed no significance(P>0.05); But in tongue cancer the"â…¡-â…¢stage"showed significantly higherthan"0-â… stage".(P<0.05)â‘¶There were 18 positive cases in 26 cases of the group with lymphonodi cervicales metastasis ,and its positive tate was up to 69.23%, which was significantly higher than the opposite group.2. The expression of TÎ’R-â…¡:â‘´There were 9 positive cases among 10 cases of normal tongue tissue, and the positive rate was 90.00%. There were16 positive cases among 40 cases of squamous cell carcinoma of tongue and the positive rate was 40.00% .In comparison with normal tongue tissue the difference was significant(P<0.05).⑵The positive expression of TÎ’R-â…¡among different age groups or different TNM stages showed no significance(P>0.05); But in tongue cancer the difference histology groups showed significant(P<0.05).â‘¶The difference between the group with lymphonodi cervicales metastasis and the opposite group was not significant (P>0.05).3. The correlation between the positive expression of TGF-β1 and TÎ’R-â…¡: There was a significant inverse correlation between loss of TÎ’R-â…¡expression and the positive expression TGF-β.Conclusions:1. The difference of the positive expression of TGF-β1 and TÎ’R-â…¡between normal tongue tissue and tongue cancer was significant .2. The positive expression of TGF-β1 correlated with lymphonodi cervicales status and TNM stage respectively ,which incorrelated with the different age groups and different histological grades.3. The positive expression of TÎ’R-â…¡correlated with different histological grades, which incorrelated with the different age groups and TNM stage respectively and lymphonodi cervicales status.4. The difference of the expression of TÎ’R-â…¡between the positive group of TGF-β1 and the negative group was significant ,and the positive expression of TGF-β1 correlated inversely with the expression of TÎ’R-â…¡. |
