Objective:To detect the expression of human papillomavirus type 16 E7 protein(HPV16 E7),transforming growth factorβ1(TGFβ1) and membrane type-1 matrix metalloproteinase(MT1-MMP) in human cervical neoplasms and to explore their possible roles in tumor formation and progression.To investigate the effects of TGFβ1 on expression of MT1-MMP in cultured Siha cells.Methods:Immunohistochemical staining technique was used to detect the expression of HPV16 E7,TGFβ1 and MT1-MMP in 64 paraffin embedded samples of different cervical lesions.Siha cells were treated with TGFβ1 (10ng/ml) for 24h.Semi-quantitative RT-PCR was performed to observe the effects of TGFβ1 on the expression of MT1-MMP.Results:With the progressing from benign to malignant lesions,the expression of TGFβ1 and MT1-MMP significantly increased(P<0.05).In HPV16 E7 positive group,the expression of TGFβ1 and MT1-MMP were higher than the HPV16 E7 negative group(P<0.01).The association between TGFβ1 and MT1-MMP expression were statistically significant(r =0.96,P<0.01).In the test group of Siha cell,the expression of MT1-MMP mRNA were higher than the control group(P<0.01). Conclusion:The expression of TGFβ1 and MT1-MMP has correlation with the development of cervical neoplasms.A higher level of TGFβ1 and MT1-MMP indicates the malignance of cervical lesions.HPV16 infection may have effects on the expression of TGFβ1 and MT1-MMP through E7 protein.TGFβ1 may up-regulate the expression of MT1-MMP in Siha cells. HPV16 E7 may have effects on the expression of MT1-MMP mediated by TGFβ1,which may contribute to cervical cancer progression.
