Purpose:(a)observing the effect of highly concentrated dextrose to the cult ure of HUVEC;(b)observing the restraining effect of kudzu root medicine to t he harm of dextrose on HUVEC.Methods:(a)Establish and make sure of cultivating HUVEC according to t he normal exterior cultivating model of HUVEC;(2) divide the normally cultiv ated HUVEC into different groups of DMEM culture fluid with dextrose densit ies(5.5mmol/L,11 mmol/L,22 mmol/L,33 mmol/L,44 mmol/L,55 mmol/L), study the effect of highly concentrated dextrose to HUVEC by cultivating res pectively 1d,2d,3d,4d and through morphology observation and MTT method; (c) divide the highly concentrated dextrose(33 mmol/L)cultivating groups into kudzu root medicine 0group,0.5mg/Lgroup,1.0 mg/L group,2.0 mg/Lgroup, 5.0 mg/Lgroup,10.0 mg/Lgroup.The normal(5.5 mmol/L) group is divided in the same way.Each group is subdivided into two groups to cultivate for 24h ours and 28 hours respectively.Use MTT method to measure the light density and calculate the restraining rate.(d) collect the cell upper clear fluid of each group after 24h and 48 hour cultivation,check the content of TM,vWF,t-PA, PAI-1 and SOD activity.The results:(a)dextrose density 22mmol/L,33mmol/L, 44mmol/L,55mmol/L cultivate 1d;11 mmol/L,22mmol/L,33mmol/L,44mmol/ L,55mmol/L cultivate 2d,3d,4d,HUVEC increase is restrained(55 mmol/L c ultivating 4dis the most obvious),which is proportional to the dextrose densit y and prolongation of time;(b) in the upper clear fluid under the condition of highly concentrated cell cultivation,TM,vWF,PAI-1 contents obviously increas e,t-Pa content and cell SOD activity obviously decrease,which is of statistical significance.The decrease of restraint rate of the kudzu root medicine group i s also of statistical significance compared with the model group;(c) the TM, vWF,PAI-1 contents in kudzu root medicine group are obviously lower than th ose in model group(P<0.01),the t-Pa content in kudzu root medicine group in obviously higher than that in model group(P<0.01),SOD activity is higher than that in model group(P<0.01).Conclusion:(a) highly concentrated dextrose has obvious restraining effec t to the increase of HUVEC and is proportional to the density and the time le ngth.(b) the kudzu root medicine can restrain the contents of TM,vWF,PAI-1, t-PA and the unusual change of SOD activity to some extent and protect th em.
