Soft Liver Eliminate Water Cataplasm Pharmaceutical Research

Ascites as a world wide disease has led thousands of people to death every year, and it has already become one of most serious diseases needed to be overcome today. However, its therapy has suffered great challenge for its regeneration, heavy adverse reaction and toxicity of drugs, bad compliance of patients as well as its high therapy cost.Recently, we found these deficiencies can be made up by TCM. Comparing the therapeutical principle, the safety, the compliance of patients and the therapy cost between the Western Medicine (WM) and the Chinese Medicine (CM), the latter can be a good supplement. In order to offer a patent drug, the study of RuanGanXiaoShui Cataplasm was carried out on the base of the clinic proven prescription from 302 hospital of PLA.The studies on extraction and segregation: First, to extract the essential oil in Cortex Cinnamomi, the CO₂ supercritical fluid extraction and steam distillation was studied, and the transferred rate was 83.3% and 70.8%, respectively. The SFE was selected for the project. Further more, to optimize the extraction parameter for the other materials, orthogonal test was used.Meanwhile, the mice transplanted with S₁₈₀ ascitic cancer model was adopted, by which the process of segregation were fixed.The studies on preparation process: The prescription and parameter were optimized with index of bio-adherence property. After drug added, the conditions were further adjusted based on the drug transdermal dynamics.The establishment of system for quality evaluation in vitro: The rolling ball test, flat traction test and 180°decoherence test were adopted to evaluate the product's viscidity and the intensity. Further more, the percutaneous absorption and the dissolution property in vitro, as well as its content determination were studied. Meanwhile, the white rabbit skin irritation of product was studied as a bio-property for evaluation. The preliminary stability experiment shows that the product was stable in 3 months. The preliminary evaluation of product's bio-activity: In order to preliminary evaluate the product's bio-activity, the mice transplanted with S₁₈₀ ascitic cancer model was adopted. Meanwhile, the volume of ascites, abdominal circumference, body weight, organ weight, and the volume of albumen in ascites and blood serum were studied. The study shows that RuanGanXiaoShui Cataplasm can reduce the volume of ascites.
