Objective:The optimum extraction process of compound Paeonia lactiflora Babu agent and the best matrix prescription of Babu agent were selected.To determine the optimal preparation process,establish the quality evaluation system and evaluate the pharmacodynamics,and provide experimental reference for the further research of the compound paeony Babu.Methods:The method of ethanol reflux extraction was used to extract the medicinal herbs of the compound Paeonia lactiflora Babu,the transfer rate of peoniflorin and cinnamic aldehyde were used as evaluation index to examine the extraction time and frequency of compound Chinese herbs and the ratio of medicinal materials,and to optimize the extraction process of medicinal materials.Taking comprehensive sensory as index,single factor method was used to investigate the type and dosage of matrix;On the basis of single factor investigation,using sensory index,initial adhesion force and sticky force as indicators,uniform design method was used to optimize the best matrix formulation of Babu agent;The content of paeoniflorin and cinnamaldehyde in the receiver were determined by HPLC,the type and dosage of the enhancers were determined;According to the provisions of the four general principles and 0122 patches of Chinese Pharmacopoeia 2015,the characters,content determination and other examinations were carried out;The initial stability of 3 groups of compound Paeoniae Alba Babu preparation for 5 months were observed by room temperature retention method;The stability of compound Paeonia lactiflora Pall Babu preparation was studied by high temperature,high humidity and strong light irradiation;A rat model of adjuvant-induced arthritis(AA)was established by injecting complete adjuvant adjuvant(FCA)into the rat's toe to investigate the effects of compound Paeonia Babu on the serum tumor necrosis factor(TNF-alpha),interleukins-1 beta(IL-1 beta)and the interleukin-6 beta(IL-6 beta)in the rat serum;Results:The best optimum extraction process:The compatibility ratio of Paeonia lactiflora and Ramulus Cinnamomi was 3:1,and the time of extraction was 1H;The best prescription of compound Paeonia lactiflora Babu:NP-700 3G,glycerin 12g,glycine aluminum 0.08g,tartaric acid 0.08g,PVP-K90 0.7G,PVPP 0.5g,2%azone,water 20g,dry extract 3.64g;A quality evaluation method for compound Paeonia lactiflora Babu preparation was established:The content of paeoniflorin in each compound Paeonia lactiflora Pall Babu agent was determined by HPLC.The content of paeoniflorin was not less than 8.18mg,and cinnamaldehyde content was not less than 0.91mg,the established method is simple,specific and reproducible;The average content of ointment was 15.03g/cm~2;The shape,heat resistance and adhesiveness accorded with the requirements prescribed in the Pharmacopoeia:The results showed that all the tests met the requirements at room temperature,indicating that the initial stability of the compound Babu agent was relatively better in 5 months;The high temperature,high humidity and strong light irradiation test of the compound Paeonia lactiflora Babu were carried out,all the tests can meet the requirements,it shows that the stability of the compound Paeoniae Paeoniae Babu agent is relatively better under the influence of various factors;The high dose group of compound paeony Babu can significantly reduce the degree of secondary side foot swelling in rats(P<0.05)and the concentration of cytokines IL-1 beta,TNF-A and IL-6 beta in the serum of rats(P<0.05);Conclusions:The preparation technology and quality control method of compound Paeonia lactiflora Babu were reliable and effective in the treatment of adjuvant arthritis rats.
