Objective:To observe Xiaxi oral liquid on spontaneously hypertensive rats(SHR) blood pressure,serum angiotensin converting enzyme(ACE) activity,cardiac and abdominal aortaⅡ-1 angiotensin receptor(AT1R) the level of expression,And the heart,blood vessels and other target organ of the protective effect and to further explore its mechanism.Methods:40 12-week-old SHR were randomly divided into SHR blank group,Xiaxi oral liquid Group,captopril group,irbesartan group,a total of four groups,each with 10, meanwhile select 10 WKY rats the same-week age were selected as comparison.①by the end of arterial pulse measurement before and after administration of 2,4,6,8 weeks the rats in blood pressure;②eight weeks after from apex organization to do abdominal aorta and slicing,HE staining method used in optical microscopy its shape changes;③eight weeks after the experiment using UV-Determination of the Group of serum ACE activity, immunohistochemistry Chemical(ICH) of rats in each group and the abdominal aorta AT1R myocardial protein expression.Results:①the treatment of rats have different levels of blood pressure drop,compared with the SHR blank,oral liquid group in Xiaxi 4 weeks after the administration significant difference(P<0.05),irbesartan group and the Cato Perindopril group from the administration 2 weeks after a very significant role in the relief(P<0.01);②rats in each group by cardiac and vascular tissue HE staining after observation,the treatment group than in blank group have varying degrees of damage mitigation;③each group of serum ACE activity:SHR blank group was significantly higher than WKY group(P<0.01),Xiaxi oral liquid group was significantly lower than the blank group(P<0.01),but still higher than the WKY group (P<0.05),irbesartan group was lower than blank group(P<0.05),significantly higher than that of WKY group(P<0.01),Captopril group dropped most significantly,compared with WKY no difference(P>0.05);④rats in each group and the abdominal aorta AT1R myocardial expression of the average value of gray:WKY group was significantly higher than the blank group(P<0.01),Xiaxi oral liquid group higher than blank group(P<0.05),lower than irbesartan group,captopril group than blank group has increased but not significantly different (P>0.05),Irbesartan group was significantly higher than the blank group(P<0.01),and WKY was no significant difference(P>0.05).Conclusions:①Xiaxi oral liquid group can significantly reduce systolic blood pressure;②Xiaxi oral liquid group is the protection of cardiac cells,vascular endothelial protection,prevention of vascular smooth muscle cell proliferation and the role of wall thickening;③Xiaxi oral liquid group to reduce serum ACE of SHR activity,with a similar mechanism of the ACEI;④Xiaxi oral liquid group can reduce the SHR cardiac and vascular expression in AT1R,with a similar mechanism of the ARB.
