Background and aim:After HGP(Human Genome Project),human genome studies gradually enters the Post genomic Era,namely gradually changes by the structural genome time to the functional genome time.Simultaneously appearance of biological chip and bioinformatics provide powerful tool for the study in Post genomic Era.In recent years,people simultaneously noted the meanings of syndrome-genome study. Regardless of expert,scholar or official,all invested the enormous attention,hoped the new breakthrough.However because syndrome-genome studies are arduous, complex,long-term,there is a little achievement.We made the pedigree expression profile as the foundation to study the deficiency-cold syndrome in the terms of signal transduction pathway.MethodWe collected 2 typical deficiency-cold syndrome pedigrees based on clinical investigation,microarrays were made to examine expression profile of deficiency-cold syndrome.Then through cluster analysis,gene function annotation, gene function enrichment analysis to mine the biological rules of deficiency-cold syndrome.After the data analysis,we take the further experiments to verify the biological foundation.ResultsThe data mining methodology of biological chip were elaborated systemically, especially the data thorough analysis method,thus providing the methodology platform for the syndrome-genome study.Through the bioinformatic analysis of expression profiles of two deficiency-cold syndrome pedigrees,especially gene function enrichment analysis,electron transport chain in mitochondrion is significant, which is the power of energy production.Through the sequencing of mtDNA regulation area,the normal person and deficiency-cold patient could be separated, which demonstrates that the mutation of mtDNA regulation area is of specificity with efficiency-cold patient. Discussion:The microarray,a kind of high flux screening method,hopefully provides a good platform for syndrome-genome study.Based on the gene function enrichment analysis, the signaling pathway are introduced into syndrome research of traditional Chinese medicine in order to be able explain the biological essence of syndrome from higher level.Through two pedigrees,4 levels data thorough analysis,electron transport chain in mitochondrion play a significant role in deficiency-cold syndrome,which explains the deficiency-cold syndrome well.But because of its complexity,it needs the further evidence to verify.
