Preliminary Evaluation Of The Turmeric Germplasm Resources And Sichuan Turmeric Good Kind Of Preliminary Screening Studies

Radix Curcumae,the root tuber of Curcuma wenyujin Y.H.Chen et C.Ling,C. longa L.and C.kwangsiensis S.G.Lee et C.F.Liang or C.phaeocaulis Val,root in the family of Zingiberaceae.The difficulty in the quality control of Radix Curcumae materials,unstable quality and significant reduction of yield as the result of their complex origin,wide distribution,multi-variety,breeding backward technology,cultivation and harvesting processing methods not standardized and the existing statutory,standards were not effective quality evaluation methods.The work in this thesis is part of the Breeding Tackling-key Project of Sichuan:Investigation on characteristics germplasm Materials and part of the technology bureau project of chengdu city:Investigation on evaluation of of Radix Curcuma germplasm and normalization of quality criteria.We mainly study the evaluation of collected 29 germplasm systematicly and good spices initial screening of chuanyujin(lvsi yujin and huangsi yujin).In the reseach on identification of germplasms,we studied on the 29 germplasms collected form the 4 provinces to 21 for plant morphology,material character,micro-character,TLC identification and HPLC fingerprint identification The study on morphous histology indicate that:germplasms collected from guangxi origined from Curcuma kwangsiensis and C.wenyujin.Deploy germacrone,curdione, curcumol and curcumine as standards for TLC identification.Established HPLC fingerprint of wen yujin,huangsi yujin and lvsi yujin,The common fingerprint of the three species can be used to identification them.According to TLC fingerprint identification and HPLC results show that similarity:There are significant differences in chemical composition and the type of content in germplasms,No.17 has a close similarity with lvsi yujin,No.19 has a close similarity with wen yujin.The conclusions is matched with the original plants,traits,microsurgery and TLC identification.In the reseach on evaluation of germplasms,we studied on the yield, cost-effective evaluation and internal quality assessment for different varieties of.yujin Determine the volatile oil,extract,the content of curcumin,established the methods to determine germacrone and curdione.Evaluation results show that:yujin collected form fujian has differences between the two larger components germplasms, the content of germacrone in germplasm of No.17 is consistent with that in peng ezhu. the content of germacrone in germplasm of No.19 is consistent with that in wen yujin,and this fingerprint match results of the evaluation.However,the germplasms of Guiyujin is promiscuity,relatively large differences in ingredients.The result is consistent with the conclusion that it can not be established the fingerprints of HPLC.From the production and chemical composition of evaluation,we found that:lvsi yujin is the best of all in production and economic benefits,particularly in those germplasms origined from Sichuan-4 conbine with the chemical composition.Huangsi yujin has the highest content of extract,volatile oil and total curcumin,the content of curcumin is significantly better than other varieties of yujin,with a difference of more than 1,000 times,particularly in those germplasms origined from Sichuan-7 has the highest content of volatile oil while particularly in those germplasms origined from Sichuan-6 has the highest content of total curcumin and extract.The highest content of germacrone and curdione-is Wenyujin,origined preferably form zhejiang-3.In view of the advantages of chuanyujin,optimization lvsi yujin and huangsi yujin for breeding germplasms.It laid the foundation for breeding of chuanyujin.In the reseach on good spices preliminary screening of chuanyujin,First of all, we screen 291 pieces of chuanyujin.Then divided those qualified sample for the A,B,C three groups according to root fresh weight and stem fresh weight of output filter.Following determine the content of total curcumine,germacrone and curdione with the A,C types.Finally,according to the final comprehensive analysis of the total chemical constituents,we screened 49 pieces of excellent chuanyujin germplasms.It included 9 pieces of lvsi yujin germplasms which are all collected form Sanqiao,and 40 pieces of huangsi yujin germplasms which are produced in Sichuan-1,Sichuan-3, Sichuan-4,Sichuan-5,Sichuan-6,Sichuan-7.The results provide the scientific theory basis for evaluation of germplasms resources,the establishment of standards and good spices initial screening of chuanyujin.
