Herpetospermum seed,a commonly used Tibetan medicinal substance,is the dried ripe seed of Herpetospermum caudigerum Wall(H. pedunculosum(Ser.) Baill.)(Cucurbitaceae),which has been recorded in Drug Standards-Tibetan Medicinal Substances(Vol.1).It is bitter in taste and cold in nature.Its actions include clearing heat from fu-organs(e.g. gallbladder),detoxifying,purging the fire,and aiding the digestion.It is indicated for liver heat syndrome,icterohepatitis,etc.To date,a few researches results on Herpetospermum seed have been reported.Yuan HL,et al reported Herpetin has been isolated from it; Zhang M,et al.isolated Herpetone(Ⅲ),dehydrodiconiferyl alcohol; Kaouadji Mourad,etc.reported it contained some benzofuran components including Herpetal,Herpetoriol;and some lignans including Herpetol,Herpetrione,Herpetetradione,Herpetetrone,and Herpepentol.The results of our early pharmacological experiments indicated that the ethyl acetate extract of the Herpetospermum seed has the effect of againsting hepatic injury.In order to find the bioactivity compounds we studied on chemical components from the ethyl acetate extract,the bioactive part for liver diseases,of the Herpetospermum seed.By means of nomal pressure silica gel column chromatography and Semipreparative High Performance Liquid Chromatography,6 compounds were isolated from it.Based on the physical and chemical fearures,and spectrograph dates,they were identified as:HerpetolideⅠ(Ⅰ),HerpetolideⅡ(Ⅱ),Herpetone(Ⅲ),dehydrodiconiferyl alcohol (Ⅳ),Pyrimidine-2,4-diol(Ⅴ),stigmasterol(Ⅵ).CompoundsⅠ,Ⅱare two new coumarin compounds and belong to a rarely knew structure mode.We named it HerpetolideⅠand HerpetolideⅡ.CompoundⅤ, Pyrimidine-2,4-diol,was isolated from the genus of Herpetospermum for the first time.Then we subjected the compoundsⅠ,Ⅲ,ⅣandⅤto vitro pharmacological experiments sequently,testing the effects of protecting the mice's liver cells when they were injured by DMSO.The results indicated the OD value of DMSO injured group,compoundsⅠgroup,and compoundⅣgroup showed significant differences compared with the control group,p<0.01;The OD value of compoundⅢgroup didn't show significant difference compared with the control group;with the DMSO group,p<0.01.The results demonstrated the DMSO could injure the hepatic parenchymal cells(compared with the control group,p<0.01). CompoundⅢcan against the hepatic cell injury induced by DMSO, increase the survival rate(compared with the control group,p<0.01).The results of our research provide the pharmaceutical substance foundation for treating the hepatic disease of Herpetospermum seed,thus will enhance the clinical use of the Herpetospermum seed and the R&D of new medicine.
