Objective:To study the effects of nourishing the Qi and remove the blood pretreatment by moxibustion on myocardial apoptosis induced by ischemia-reperfusion.Methods:The left anterior descene coronary artery was ligated to establish the ischemia-reperfusion heart model in rat.Immunohistochemical staining was used to detect the changes of apoptosis and protein expression of caspase-3,bcl-2,bax,fas,TNF-αand NF-kB.Histopathological changes in myocardium was also observed.Result:(1) The number and size of cardiomyocyte cell reduced after ischemia-reperfusion and increased by nourishing the Qi and remove the blood pretreatment of moxibustion.(2) The expression of caspase-3 gene increased by ischemia-reperfusion and decreased by nourishing the Qi and remove the blood pretreatment of moxibustion.(3) The expression of bcl-2 gene decreased by ischemia - reperfusion and increased by nourishing the Qi and remove the blood pretreatment of moxibustion.(4) The expression of bax and fas gene inregulating after schemia-reper fusion and downregulating after nourishing the Qi and remove the blood pretreatment of moxibustion.(5) The expression of TNF-αand NF-kB increased by ischemiareperfusion and decreased by nourishing the Qi and remove the blood pretreatment of moxibustion.Conclusions:Nourishing the Qi and remove the blood pretreatment by moxibustion had obviously preventive and therapeutic effects on reperfusion injury and could inhibit cardiomyocyte apoptosis.The mechanism might be that bcl-2 gene expression was increased,caspase-3 gene,bax gene,faa gene,TNF-αand NF-kB were decreased following ischemia and reperfusion.
