ObjectiveThrough establishing the rabbit injury model of myocardial ischemia-reperfusion, to observe the effection of Electroacupuncture Pretreatment (EP) on the expression of Fas/FasL and Formazan,and the Influence of apoptotic index, comparing the effects of EA at different acupoints(Neiguan(PC6),Lieque (LU7),and Hegu(LI4)); discussing the Protective Function of EP on MIRI; confirming the specific correlation between the heart with the pericardium channel.Methods40New Zealand big-ear rabbits were randomly divided into5groups:control group A (sham-operation), group B(ischemia reperfusion), group C(EP of Neiguan(PC6)), group D(EP of Lieque (LU7))and group E(EP of Hegu(LI4)),8in each group. Group A and B have no need to treat with EP. Group C、D and E were treated with EP before Establishing method.Establishing myocardial ischemia-reperfusion injury model with the method of Ligating the anterior descending branch of the left coronary artery for30mins and then reperfusing for60mins. After the last bunding, all groups were opened chest to draw materials.Group A were threaded and standed for30min,then pulled out the line and continue to stand for60minutes,then drawwed materials.Group B, C,D and E were conducted ischemia-reperfusion, then drawwed materials. The expression of Fas/FasL was determined by immunohistochemical method;the apoptosis was determined by TUNEL; the Formazan was determined by MTT; ultrastructure was observed by electron microscopy.Results1. Ultrastructure changes of the ischemic tissue in left ventricular of MIRI rabbits was observed by electron microscopy.The structure of Group A and C is more normal, Group A is better than Group C.B,Dand E lost their normal structure.2. Fas/FasL expression comparison of each group in rabbit myocardial tissue.3.Fas and FasL expression of group A is almost normal; Fas and FasL of Group B,D and E were increased significantly; Fas and FasL expression of group C was reduced significantly than group B,D and E. 4.Comparison of AI of each group in rabbit myocardial tissue:The AI of group A is almost normal. AI of Group B,D and E were increased significantly;compared with group B,D and E, the AI of group A was reduced significantly, and no significant differences with group A.4. Comparison of Formazan of each group in rabbit myocardial tissue:The Formazan of group A is almost normal. Formazan of group B,D and E were reduced significantly;compared with group B,D and E, Formazan of group C was increased significantly.Conclusion:1. EP Neiguan(PC6) can decrease the release of Fas, and FasL,and keep the high level of Formazan;2.The intervention effects of Group C is better than group D and E, the specific correlation between the heart with the pericardium channel.
