Objective:+33371A/G polymorphism of the AdipoR2 gene was detected in the test people of the 100 NAFLD and 100 controls by physical test center of the Third Hosipital of Datong Shanxi Province,to explore the relationship between NAFLD and polymorphism in the locus in the groups.Methods:We collected 100 NAFLD and 100 controls,as a study,measured the height,weight,waist circumference,hip circumference,blood pressure etc.of these patients and the controls,We collected blood specimens of these patients and the controls to test the blood glucose,blood lipids,liver function,hepatitis markers,insulin of them.We extracted genomic DNA of blood samples and adopted method of PCR-RFLP to detect+33371A/G polymorphism of the AdipoR2 gene of NAFLD patients and controls.Then we analysed the clinical data.Results:1.BMI,TG,LDL-C,waist circumference,WHR,BP,FPG,FINS,HOMA-IR and ALT,AST, UA in the NAFLD group was higher than the controls,HDL-C in the NAFLD group was lower than the control group.It has significant difference(P<0.01),NAFLD group has the charactics of high TG hyperlipidemia and low HDL-C hyperlipidemia.2.The PCR products after digestion displayed 3 genotypes,including AA,AG and GG.The frepuencies of AA,AG and GG genotypes in NAFLD group(no.=100)and control group (no.=100) were 0.34,0.44,0.22 and 0.17,0.55,0.28.The A allelic frequency is 56%and 44.5%in NAFLD and controls.Respectively,there was a significiant difference in genotypic and allelic frequencies distribution of +33371A/G polymorphism of the AdipoR2 gene between NAFLD patients and controls subjects(P<0.05).3.Make the clinical indicators as the study and re-grouped by genotype.It showed that BMI,TG, FINS and HOMA-IR with AA+AG genotype were higher than with GG genotype,and it has significant difference(P<0.05).4.We compared clinical data in AA+AG genotype with in GG genotype of the NAFLD group. It showed ALT in the AA+AG genotype is higher than in the GG genotype,and higher than normal,and it has significant difference(P<0.05).High ALT(45 cases) is more than normal ALT(33 cases) in AA+AG genotype carriers.There is significant difference(P<0.05).The possibility of ALT increased in AA+AG genotype carriers is 5.925 times of GG genotype carriers in NAFLD group(OR = 5.925 P<0.01 95%C1(1.898~20.000)). Conclusion:1.NAFLD patients is closely associated with metabolic syndrome.We should put emphasis on screening and controlling NAFLD.It has important significance on the prevention of type2Diabetes,metabolic syndrome,and prediction of the incidence of cardiovascular events.2.Carrying A allele increased the risk of suffering from NAFLD ofAdipoR2 gene +33371 in the these people.A allele may be associated with BMI,TG,FINS and HOMA-IR.3.High ALT with AA+AG genotype is more than normal ALT in the NAFLD patients,(P<0.05),which indicated that the polymorphism probably plays an role in the occurrence of liver inflammation and development.
